The current study aimed to evaluate the relationship between seminal plasma proteins and sperm quality in Toraya buffalo bulls. Semen samples were obtained from eight Toraya buffalo bulls aged 4-10 years. Semen collection was conducted using an artificial vagina, followed by assessment through macroscopic and microscopic analysis. The concentration of seminal plasma protein was measured utilizing the bicinchoninic acid (BCA) protein assay, followed by protein analysis through 1D SDS-PAGE based on protein molecular weight. The sperm quality of Toraya buffaloes showed an average semen volume of 2.08±1.12mL, sperm motility of 73.87±5.30%, sperm concentration of 883.12±381.35×10 6 /mL, viability of 81.47±3.79%, abnormality of 7.36±3.24%, membrane integrity of 83.14±5.19%, and acrosome integrity of 92.98±2.56%. The parameter intact acrosome in sperm quality showed a positive correlation (0.73) with sperm viability with a significant (P<0.05). The total motile sperm per ejaculate was 1326.61±714.99x10 6 , and the total straw per ejaculate was 54.06±28.59. The average seminal plasma protein concentration in Toraya buffalo was 161.41±12.41µg/mL. The Pearson correlation results of seminal plasma protein correlated with intact acrosomes with a significant P<0.05. The analysis of seminal plasma protein bands using 1D SDS-PAGE founded the presence of 12-19 protein bands with molecular weight ranging from11-155kDa. This study concluded a significant correlation between the seminal plasma protein concentration and sperm acrosome integrity in Toraya buffalo bulls. The identification of 12-19 protein bands that correlate with these factors is a promising finding and can be utilized to determine the reproductive quality and fertility of the bulls.

This study aimed to determine the morphometric performance of polled Bali cattle aged 12-15 months and the influence of the cows on the morphometrics of breeding cattle.The research material consisted of 120 head Bali cattle divided into 40 head polled cattle, 40 head with horns, and 40 cows with horns. The study was conducted in Barru District, South Sulawesi Province. The study parameters consisted of withers height, body length, and heart girth. The data were statistically analyzed using the independent sample t-test, and the effect of the morphometrics of the cows on the calves was analyzed using simple regression analysis. Measurement data were analyzed using the IBM SPSS Statistics application. The results showed a very significant difference in the body length of polled and horned males (p<0.01). Significant differences were also found in withers height and body length between polled and horned female calves (p<0.01). Polled Bali cattle aged 12-15 months have a larger morphometric size than those with horns. It is hoped that identifying the superiority of polled Bali cattle will continue at different age statuses to compile information as material for the Indonesian National Standard for hornless Bali cattle.

Background and Aim: Fertility is crucial for enhancing the efficiency of livestock production, as it directly impacts the reproductive rates. A comprehensive understanding of the relationship between sperm quality and embryo development is key to optimizing reproductive outcomes and improving the quality of livestock. This study analyzed the developmental competence of in vitro embryos recovered from Bali cattle with normal or poor sperm motility. Materials and Methods: Nine bulls with normal fresh semen (NFS) or poor fresh semen (PFS) motility were ejaculated for semen. Semen ejaculates, including volume, motility, and sperm concentration, were evaluated immediately after collection to measure the quality of the fresh semen. Frozen semen was evaluated using computer-assisted semen analysis (CASA) for motility, progressive sperm motility, distance curve path, distance curve linear, distance straight line, average path velocity, curvilinear velocity, linear velocity, straightness (STR), linearity of forward progression (LIN), wobble, and average lateral head displacement (ALH). Bull groups were used to determine in vitro embryo cleavage ability after fertilization of Bali cattle. Ovaries of Bali cattle were collected by slicing, and only cytoplasmic oocytes with compact cumulus cells were used in this study. The oocytes were matured, and in vitro fertilization was performed using fertilization media with a final sperm concentration of 1.5 Ã— 106 spermatozoa/mL. After 48 h, the embryo cleavage ability of the cultured oocytes was evaluated. Results: There were significant differences in motility values between the NFS and PFS groups; however, there were no significant differences in the volume or sperm concentration. There was a significant difference in the LIN value between the groups but no significant differences in other CASA parameters. There were no significant differences in the cleavage rate and morula between the groups, but a positive correlation was observed between the cleavage rate and the morula and between the morula and ALH. A significant negative correlation was observed between the cleavage rate and STR and between the morula and STR; no significant differences were observed for other variables. Conclusion: Despite variations in sperm characteristics, both normal and poor sperm motility demonstrated comparable in vitro embryonic development competence. These findings provide important insights into the fertility potential of Bali bulls, providing valuable information that can enhance selection strategies to improve the quality of livestock production.
