Sphingomonas changnyeongensis sp. nov. isolated from the Hapcheon–Changnyeong barrage area in the Nakdong river

Abstract: The novel bacterial strain C33T was isolated from a freshwater sample collected from the Hapcheon–Changnyeong barrage. The Gram-negative, motile, yellow-pigmented strain C33T was characterized as a rod-shaped and strictly aerobic bacterium. A 16S-rRNA phylogenetic analysis revealed that this strain was most closely related to Sphingomonas changbaiensis V2M44T, Sphingomonas taba… Show more

“…S4). Isoprenoid quinones were examined by isocratic high-performance liquid chromatography using Nova-Pak C18 reverse-phase columns (particle size, 4 µm; Waters) [33, 34]. The mobile phase consisted of a methanol–isopropyl ether solution (3 : 1; v/v) flowing at a rate of 0.7 ml min −1 .…”

“…To identify the bacterium, freshwater samples collected from HCW (35° 35′ 32.5″ N 128° 21′ 25.4″ E) were filtered through a 22 µm filter paper. The samples were then serially diluted and spread on Reasoner's 2A (R2A) agar (0.05 g yeast extract, 0.05 g proteose peptone No.3, 0.05 g casamino acid, 0.05 g d -glucose, 0.05 g soluble starch, 0.03 g sodium pyruvate, 0.03 g K 2 HPO 4 , 0.005 g MgSO 4 and 1.5 g agar powder in 100 ml distilled water, pH 7) plates and incubated for 3 days at 30 °C [14]. Strain HC41 T was isolated from one of these plates.…”

“…The genus Sphingomonas was originally described by Yabuuchi et al [1] and then emended by Busse et al [2] and Chen et al [3]. At the time of writing, the genus Sphingomonas consists of more than 190 recognized species (www.bacterio.net), including the recently described species Sphingomonas aliaeris [4], Sphingomonas sabuli [5] and Sphingomonas changnyeongensis [6]. Members of the genus Sphingomonas are Gram-negative, yellow-pigmented, aerobic, non-fermentative, non-sporeforming, non-motile or motile rods and are characterized chemotaxonomically by the presence of ubiquinone Q-10 and 2-hydroxy fatty acids [7, 8].…”
