Spi-B is critical for plasmacytoid dendritic cell function and development

Abstract: Plasmacytoid dendritic cells (pDCs), originating from hematopoietic progenitor cells in the BM, are a unique dendritic cell subset that can produce large amounts of type I IFNs by signaling through the nucleic acid-sensing TLR7 and TLR9 (TLR7/9). The molecular mechanisms for pDC function and development remain largely unknown. In the present study, we focused on an Ets family transcription factor, Spi-B, that is highly expressed in pDCs. Spi-B could transactivate the type I IFN promoters in synergy with IFN re… Show more

“…Cluster 33 is enriched in plasmacytoid dendritic cells (pDC) and contains TLR9, known to be pDC-restricted in humans, and the pDC transcription factors IRF4 and SPIB [50], again consistent with previous mouse data [2]. It is not our purpose to annotate all of the clusters processively.…”

“…pDC numbers are reduced in the bone marrow of Spib −/− mice but are increased in the periphery, suggesting impaired retention in the bone marrow 59 . Furthermore, Spib −/− pDCs exhibit attenuated expression of pDC-specific genes and display defects in type I IFN production following TLR7 or TLR9 stimulation.…”

“…However, Spi-B does not express in lymphoid cells (neuthrophils and monocytes) (Chen et al, 1995) because it prevents differentiation of hematopoietic progenitor cells toward the lymphoid lineage (Schotte et al, 2003). Spi-B is critical for development and function of both mouse (Sasaki et al, 2012) and human (Schotte et al, 2004) pDCs. E2-2 and Spi-B cooperate in stimulating pDC differentiation in order to overcome Id2 enforced block in pDC development (Nagasawa et al, 2008).…”

“…E2-2 and Spi-B cooperate in stimulating pDC differentiation in order to overcome Id2 enforced block in pDC development (Nagasawa et al, 2008). In synergy with IRF7, Spi-B activates expression of IFN-I in pDCs (Sasaki et al, 2012). Finally, Spi-B supports survival of pDCs and their precursors by up-regulating expression of anti-apoptotic protein Bcl2-A1 (Karrich et al, 2012).…”
