Spontaneous deletions and flanking regions of the chromosomally inherited hemolysin determinant of an Escherichia coli O6 strain

Abstract: The hemolytic Escherichia coli strain 536 (06) propagates spontaneous hemolysin-negative mutants at relatively high rates (10-3 to 10-4). One type of mutant (type I) lacks both secreted (external) and periplasmic (internal) hemolysin activity (Hlyej-/Hlyjn-) and in addition shows no mannose-resistant hemagglutination (Mrh-), whereas the other type (type II) is Hlyej-/Hlyjn' and Mrh+. “…We then subcloned the 5.3-kb BamHI-EcoRI fragment, which spans the region 0.2 kb left of P10 to the middle of P8 (Fig. 4), into pBR322 (pANN801-11 13. Transformation of E. coli K-12 HB101 cells with this plasmid resulted in the production of fimbriae and mannoseresistant hemagglutination.…”

“…Isolation of chromosomal and plasmid DNA. Chromosomal DNA was isolated as described previously (13). Plasmid DNA from clones carrying recombinant DNA was screened by the cleared lysate procedure (4), and preparative DNA isolation was achieved as described previously (11).…”

“…The first intensively studied Pais were those which are part of the genomes of uropathogenic and enteropathogenic E. coli strains (Hacker et a/., 1983;Low et a/., 1984;Muhldorfer and Hacker, 1994;McDaniels et a/., 1995;Blum et a/., 1994;Swenson et a/., 1996). The uropathogenic E. coli strains 536 (06:K15) and J96 (04:K6) carry Pais with sizes of 70-190 kb (see Table I), The Pais of strain 536, indicated in Fig.…”

“…E. coli CFT073 was isolated from the blood and urine of a hospitalized patient with acute pyelonephritis and urosepsis (78) and E. coli 536 was isolated from a patient with pyelonephritis (79). Transurethral infections with CFT073 or 536 were performed in mice immunized with vaccine formulations containing Hma, IreA, IutA, or FyuA.…”
