Stability of Sulfonamides, Nitrofurans, and Chloramphenicol Residues in Preserved Raw Milk Samples Measured by Liquid Chromatography

Abstract: A stability study was made of 10 antimicrobials: 6 sulfonamides, 3 nitrofurans, and chloramphenicol residues in raw milk samples preserved with 0.1% potassium dichromate (K2Cr2O7) and 0.05% mercuric bichloride (HgCl2) during cold storage for 7 days. Preserved milk samples fortified with 50 ppb of each antimicrobial were analyzed by liquid chromatography (modified AOAC Method 993.32). Drugs were extracted with chloroform–acetone after solvent evaporation residues were dissolved with aqueous sodium acetate buffe… Show more

“…In a study on ultrafiltration removal of the SDM residues, a discontinuous diafiltration at 55C, using 10,000 Da molecular cutoff membrane, significantly removed the residues from artificially contaminated milk with reduced residue levels both in concentrates and retentates (Higuera Ciapara et al 1997). In another study on stability of 10 antimicrobials in raw milk preserved with potassium dichromate or mercuric bichloride at cold storage for 7 days, a significant loss of 26.2% of SDM residue was reported in the case of only mercuric chloride preservation (Noa et al 2002).…”

“…The paper's focus on sulfonamide/trimethoprim resistance is not unreasonable even though these antimicrobials have not been used in Chilean aquaculture since 2009, and when they were, accounted for only 0.01-0.02% of the antimicrobials used for this purpose (Domínguez et al, 2019). Sulfonamides/trimethoprim, like quinolones, have a potentially long half-life in the environment (Noa et al, 2002;Białk-Bieli nska et al, 2012), so that the geographically concentrated use of even these small amounts could result in their residues still being present in the aquatic environments from where the authors obtained their isolates. In 2007, for example, close to 1,000 tonnes of antimicrobials were used in Chilean aquaculture (Cabello et al, 2013).…”

“…Sulfonamides (SAs) are broad-spectrum antimicrobials inhibiting both Gram-positive and Gram-negative bacteria, as well as some protozoa, which are administered orally or mixed with animal feed of meat-producing animals to prevent and control a great variety of bacterial diseases [ 14 , 19 ]. However, the widespread use of SAs in veterinary medicine may lead to SA accumulation in foodstuffs, which probably causes a variety of untoward reactions to humans after intake of animal-derived food products.…”
