Stability of the X-ray contrast agent iodixanol=3,3',5,5'-tetrakis(2,3-dihydroxypropylcarbamoyl)-2,2',4,4',6,6'-hexaiodo-N,N'-(2-hydroxypropane-1,3-diyl)-diacetanilide towards acid, base, oxygen, heat and light

Abstract: Even under highly acidic and basic conditions, iodixanol is stable.

“…where Hf(B) is the heat of formation for compound B. A position with high proton affinity (PA) is easier protonated than a position with lower PA. As earlier shown for iodixanol, [28] the proton affinities for the present iodinated compounds are highest for the carbonyl groups, lower for nitrogen atoms and lowest for hydroxyl groups and aromatic carbon atoms. Among aromatic carbon atoms, the iodinated positions have the highest proton affinity and their protonation increases the C-I bond length.…”

“…31 Lowviscosity, iodine-based in vivo contrast agents are metabolized quickly, creating unstable images that fade rapidly from within the capillary lumina. [40][41][42] Reconstructing images of the whole vascular tree system at once gives rise to further problems. 43,44 Intensity thresholding inhibits reconstruction from clearly contrasting lumen-filled capillaries from their surrounding tissues, and at the same time demonstrates the larger vasculature.…”
