Staining intensity of individual osteons correlated with elastic properties and degrees of mineralization

Abstract: Surface staining is widely used for histological studies involving undecalcified thick bone sections. Individual osteons, particularly newly formed ones stained with toluidine blue (TB), show various color intensities. We studied the correlations between TB color intensity and the differences in stiffness and degree of mineralization of individual osteons in undecalcified histological sections of goat tibial diaphysis, measured by scanning acoustic microscopy (SAM) and contact microradiography (CMR), respectiv… Show more

“…8a) [110]. The method was controlled by SAM and microradiography and the staining intensity negatively correlates with the degree of mineralization [111]. The staining does not work on thin microtome sections, and it is likely that the large amount of the poly-methylmethacrylate remaining in polished sections limits the access of the stain since decalcified bone embedded similarly is uniformly and deeply stained.…”

“…b Quantification of callus area. c Longitudinal follow up of fracture healing from week 0 to week 6, with larger callus size observed in the KO mice compared with the WT mice at 2, 3, and dehydrated in graded concentrations of ethanol and embedded without decalcification in the modified methyl methacrylate (MMA) using our previously established protocol [20]. Briefly, the femur was dehydrated in 70%, 85%, 90%, and 100% graded ethanol and then submerged in xylene for three times.…”

“…28 According to Donath's method, 29 the embedded osseous specimens were sectioned in longitudinal direction parallel to the long axis of the micro-screw by a section cutter (SP 1600, Leica Instruments, Nussloch, Germany) at a low speed (0.4 mm/min). Three to four hard tissue sections, 60-to 80-mm thick ( Figure 2B,C), with the micro-screw's long axis in the force vector plane, were obtained from each micro-screw.…”
