Standardization ofin vitromethods

Abstract: IgE-antibody analysis is a major diagnostic procedure and a primary tool in allergological research. The determination of sensitization frequencies and antibody concentrations against allergens of defined sources provides critical information for the estimation of the relative importance of food and environment in clinical allergy. True quantitation is essential and requires assay designs providing allergen excess and mass unit calibration. Standardized and reproducible methods show geographic and culture depe… Show more

“…During this time, there have been continued technical improvements, including the use of a well-defined calibration linked to the World Health Organization-determined IgE standard, well-standardized allergen extracts to bind all antibodies without competition between classes, and absolute linearity of the test at different concentrations of IgE antibodies. 25 This has led to newer tests that can measure the IgE antibodies with very high precision in quantitative terms. 26,27 With respect to the assessment of sensitization status, the important point practicing physicians need to consider is how much sensitization contributes to the symptoms.…”

“…Measurements of specific IgE binding were performed by using the ImmunoCAP assay platform (Phadia, Uppsala, Sweden). 20,21 Results are reported quantitatively using a kilounits per liter scale (kU/L). The calibrator is IgE bound to anti-IgE by using a 6-point quantitative curve.…”

“…The skin‐prick test was considered positive when the weal diameter was ≥3 mm. For the specific IgE 0.35 kU/l was used as cutoff level (4). The chi‐squared test was used for the analysis of data.…”
