Standardized Extract of Valeriana officinalis Improves Overall Sleep Quality in Human Subjects with Sleep Complaints: A Randomized, Double-Blind, Placebo-Controlled, Clinical Study

Harshith Chandra Shekhar,
Lincy Joshua,
Jestin V. Thomas

Abstract: Introduction: Sleep deficit or poor sleep leads to ill-health, whereas sleep deprivation for longer periods of time increases the risk of developing adverse conditions associated with poor quality of life, and high socioeconomic impact. The treatments for sleep disturbances include melatonin and over-the-counter medicines like diphenhydramine and doxylamine, all of which have negative side effects. Valerian (Valeriana officinalis L.) is a traditional herb and the most preferred alternate sleep solution to mana… Show more

“…The spray-dried powder form of valerian extract was used, containing a hydro-alcoholic extract of valerian roots (OmniActive Health Technologies, Mumbai, India) at 26.23%, hydroxypropyl methylcellulose at 73.27% (Novo Excipients, Navi Mumbai, India), and colloidal silicon dioxide at 0.5% (Daksh Medicare, Mumbai, India). The composition was finalized with a 2% valerenic acid content, as determined through HPLC analysis [ 23 ].…”

“…This research aimed to verify the impact of a novel valerinic acid (VA, Pdr%2) extract on sleep quality and its molecular action mechanisms by utilizing models of sleep induced by pentobarbital and caffeine in mice. This formula is considered a promising therapeutic option because it is affordable and has not shown any documented adverse side effects compared to other compounds [ 23 ]. Additionally, the study evaluated the serum levels of serotonin, melatonin, dopamine, malondialdehyde (MDA), the activity of antioxidant enzymes (SOD, CAT, and GPx), and the molecular effectiveness of the GABA and NMDA receptors, alongside apoptotic indicators Bax, Bcl2, and caspase-3.…”

“…Among the traditional medicinal plants with anxiolytic effects used worldwide, two species deserve attention: Valeriana officinalis and Piper methysticum [7]. Clinical trials evidenced the ethnopharmacological anxiolytic effects of V. officinalis and P. methysticum [8,9]. Phytotherapeutics from these species are commercialized as mild anxiolytics.…”
