STAT5 interferes with PD-1 transcriptional activation and affects CD8+ T-cell sensitivity to PD-1-dependent immunoregulation

Abstract: Programmed death-1 (PD-1) is a co-inhibitory receptor that dampens immune responses upon the interaction with PD-L1 and PD-L2. Although the PD-1 expression on T cells is known to be activation-dependent, how cytokines modify its regulation is not fully resolved. Using polyclonal T cell activation to study the cytokine-dependent PD-1 regulation, we found that IL-2 inhibited transcriptional upregulation of PD-1 despite the promotion of T cell activation. IL-2-mediated reduction in PD-1 expression augmented CD8 +… Show more

“…The deletion of MCT1, a transporter of LA, 166,167 plays a pivotal role for tumor‐infiltrating Tregs to uptake lactate to inhibit tumor growth and enhance immunotherapy response 168 . In addition, NFAT1 positively regulates the expression of PD‐1 on T cells 169–172 . Therefore, molecular‐targeted immunotherapy against LA provides a novel insight for the clinic.…”

“…Besides potent T cell growth factor activity, IL-2 can augment CD8 T cell activities against PD-L1-expressing target cells by reducing PD-1 expression [ 22 ]. During the infusion phase of our treatment, IL-2 removal led to increased safety but resulted in a loss of PD-1 inhibition.…”
