Statistical optimization of production conditions of β-glucosidase from Bacillus stratosphericus strain SG9

Abstract: The present study illustrates the optimization and characterization of β-glucosidase from a bacterial isolate, strain SG9. Sixty-eight different variables were first screened by one factor at a time method. The screened variable optimization was then performed by Plackett-Burman design followed by Box-Behnken response surface methodology. Thirty-one variables were screened, of which five variables were found to be significant. Box-Behnken design was then performed using the most significant variables, viz., es… Show more

“…β-Glucosidases are widely applied in the food, animal feed, pharmacy, and textile industries. Several Bacillus species, such as B. subtilis ( 17 ) and B. stratosphericus ( 18 ), have been found to secrete β-glucosidases, but the enzyme yield still needs to be improved. The fungus Trichoderma reesei was also reported to produce β-glucosidase but in very small amounts ( 19 ).…”

“…When In this investigation, PBD and RSM were employed to identify the critical factors affecting β-glucosidase production by C. rhodanensis DK. Numerous research reports have documented the application of these methods for medium optimization with various microorganims, including Zobellella denitrificans [74], A. versicolor [75] and B. stratosphericus [76]. Based on the findings from PBD, it was evident that among the six component factors considered, yeast extract, peptone and xylose/xylan had a statistically significant effect on enzyme production.…”

“…Moreover, the result of the degree of freedom (DF) is 9 which is equivalent to the independent observation number. The p value can be taken as factor for significant verification of each coefficient which explains the interaction between the strength of each parameter and mutual interaction pattern between variables [30]. Several criteria were used to check goodness of fit of the model.…”

“…The determination coefficient (R 2 ) value 0.9372 indicates that the model was able to explain 93.72% of the variability in the response. In addition, the value of R 2 also is presented a correlation between the predicted and the experimental values [30], [31]. Also the adjusted determination coefficient (adj.…”
