Conventional liposome preparation methods bear many limitations, such as poor entrapment efficiencies for hydrophilic drugs, batch size limitations, and limited options for aseptic manufacturing. Liposome preparation by dual centrifugation (DC) is able to overcome most of these limitations. DC differs from normal centrifugation by an additional rotation of the samples during the centrifugation process. Thus, the direction of the centrifugal forces changes continuously in the sample vials. The consequential powerful sample movements inside the vials result in powerful homogenization of the sample. Since this "in-vial" homogenization is optimal for viscous samples, semisolid "vesicular phospholipid gels" (VPGs) are preferred intermediates in the liposome manufacturing by DC. The DC method easily enables aseptic preparation and is gentler as compared to other methods, such as high-pressure homogenization. The method allows very small samples to be prepared, and VPG batches down to 1-5 mg scale have been prepared successfully. VPGs have several applications; they are attractive as depot formulations, or as stable storage intermediates, and can be easily transferred into conventional liposomal formulations by simple dilution. Here, we aim to present the novel DC-liposome technique; the concept, advantages, and limitations; and provide an overview of the experiences of liposome preparation by DC so far.

“…Platelets (10e 6 /mL) incubated with free CF 0.3 nmol/mL gave a fluorescence intensity level 17 times less than when they were incubated with CF-liposomes containing the same amount of encapsulated marker (see Figure 1a). This shows that in the platelet-CFliposome mixture, the hydrophilic marker contained in the aqueous core of the liposomes (Tardi et al, 2001) becomes associated with the platelet, while the free marker does not. Washing the platelets after incubation with their liposomes did not significantly alter the amount of platelet fluorescence nor did the presence of EDTA, or pre-incubation of the platelets with a metabolic poison cocktail.…”

To clarify the interactions of liposomes with blood cells, this study examined the behaviour of liposomes of a range of compositions in the presence of purified human blood cells in buffer or plasma; or in whole blood, or in mice in vivo. Liposomes, labeled with the hydrophilic fluorochrome, carboxy fluorescein (CF), or with membrane-sequestering R18 or FITC-labeled phospholipids, were mixed with blood cells and the appearance of the fluorochromes in the blood cell population was monitored by flow cytometry. Irrespective of composition, with or without poly(ethylene glycol), all types of liposomes were found to interact rapidly and dose-dependently with red cells, leukocytes and platelets, both in vitro and in vivo. This took place equally in the presence and the absence of plasma proteins and functional enzyme cascades, suggesting that the prime facie interaction is opsonization-independent and is consistent with liposome-blood cell fusion.

The objectives of this study were to investigate the influence of gamma irradiation for sterilization on poly(D,L-lactide-co-glycolide) (PLGA) with different molecular weight and the effect of gamma irradiation on the release behavior of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU, carmustine) from PLGA wafer with various irradiation doses. The effect of gamma irradiation on PLGA was evaluated by gel permeation chromatography (GPC), differential scanning calorimetry (DSC), and electron paramagnetic resonance (EPR). The weight average molecular weight (M w ) and glass transition temperature (T g ) of PLGA decreased after gamma irradiation. The extent of M w reduction was dependent on irradiation dose and PLGA molecular weight. Using EPR spectroscopy, we successfully detected gamma irradiation induced free radicals in PLGA. The gamma irradiation increased the release rate of BCNU from PLGA wafer at applied irradiation doses except 2.5 Mrad of irradiation dose in this study.
