Steatogenesis in adult-onset type II citrullinemia is associated with down-regulation of PPARα

Abstract: SLC25A13 (citrin or aspartate-glutamate carrier 2) is located in the mitochondrial membrane in the liver and its genetic deficiency causes adult-onset type II citrullinemia (CTLN2). CTLN2 is one of the urea cycle disorders characterized by sudden-onset hyperammonemia due to reduced argininosuccinate synthase activity. This disorder is frequently accompanied with hepatosteatosis in the absence of obesity and ethanol consumption. However, the precise mechanism of steatogenesis remains unclear. The expression of … Show more

“…Diabetes mellitus (DM) was judged as fasting blood sugar (FBS) >120 mg/dL or hemoglobin A1c (HbA1c) >6.5% after conversion into National Glycohemoglobin Standardization Program values or the use of glucose‐lowering drugs . Hyperlipidemia (HL) was diagnosed when serum total cholesterol (T‐chol) was >220 mg/dL or triglycerides (TG) were >150 mg/dL in a fasting state or the use of lipid‐lowering agents …”

“…Although, sometimes, simple steatosis is alluded to as 'nonalcoholic fatty liver' it is, nevertheless, a disease which is worth treating not only to untrigger its potential progression to NASH but also to reduce the risk of [128]. Consistently, PPAR-a activation improves hepatic IR and steatosis in high-fructose-diet-fed mice [129].…”

“…Preparation of whole liver homogenates was conducted as described previously [15–17] and their protein concentrations were measured colorimetrically with the BCA™ protein assay kit. Whole liver homogenates (30–80 μg of protein in each lane) were separated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis.…”
