Steroidogenic enzyme P450c17 is expressed in the embryonic central nervous system

Abstract: Neurosteroids are steroids that are synthesized de novo in the brain and include some classical (adrenal and gonadal steroids) and some unique brain-specific steroids. Neurosteroids are thought to mediate their action through ion gated channel receptors such as gamma-aminobutyric acid(A) and N-methyl-D-aspartate rather than through classical nuclear steroid hormone receptors. Some enzymes involved in neurosteroidogenesis have been identified as those found in steroidogenic tissues, and some may be unique to th… Show more

“…The finding that both enzymes were expressed strongly in the fetal brain and adrenal gland is consistent with our previous observations (Nguyen et al 2003a, c) and studies in rodents by others (Voutilainen & Miller 1986, Compagnone et al 1995, Narasaka et al 2001. Expression of P450 scc was significantly decreased in the adrenal gland after hypophysectomy, consistent with the stimulatory role of ACTH in the regulation of adrenal expression of P450 scc and production of pregnenolone in the fetus (Simmonds et al 2001).…”

“…P450c17, the enzyme that is required for synthesis of DHEA from pregnenolone, is found in specific neurons of embryonic rodent brains (Compagnone et al, 1995b). P450c17 expression is mainly neuronal; its expression can be found as early as embryonic day 9.5, and persists in the central nervous system during development.…”

“…Male and female affected infants lack cortisol production and are hypertensive. To determine the location of P450c17 expression during early embryonic development in wild-type mice, we performed timed matings of superovulated female mice, obtained E7.0 embryos, and analyzed these embryos by immunocytochemistry with an antibody to human P450c17 that readily and specifically detects mouse P450c17 (12). P450c17 was detected in the embryonic endoderm of E7.0 embryos (Fig.…”

“…P450c17 is also expressed in the fetal mouse brain beginning at embryonic day 9.5 (E9.5) (12). At this time, P450c17 is found in cells migrating from the neural crest, and subsequently, P450c17 is found in many cells derived from the neural crest.…”
