Stigmasterol Alleviates Cutaneous Allergic Responses in Rodents

Abstract: The therapeutic potential of stigmasterol, a natural steroid alcohol with established immune-modulatory properties, was assessed on allergic cutaneous responses. We examined its suppressive effect on immunoglobulin E (IgE)-mediated active cutaneous anaphylaxis (ACA), compound 48/80 (C48/80)-induced pruritus, and irritant dermatitis induced by 12-O-tetradecanoylphorbol-13-acetate (TPA). Stigmasterol at 10–100 mg/kg significantly inhibited ACA with reduction in reaction area and concentration of the extravasated… Show more

“…From subfraction BC-3-2, with 27.8 % of edema inhibition, we identified, by direct comparison with an authentic sample available in our laboratory, the phytosterol stigmasterol as the main component which is already known for its anti-inflammatory effect in several models. There are many reports of the anti-inflammatory activity of this compound, and its high percentage of reduction of edema formation induced by TPA in mice, which is associated with a reduction in neutrophil infiltration into inflamed tissues has been described [23]. These results allowed us to attribute the anti-inflammatory activity of leaves of B. copallifera to the presence of α-amyrin (3), 3-epilupeol (4) and stigmasterol (5).…”

“…In one of the pairwise comparisons, 121 compounds were shared between the 2 (one Kampo and one TCM) formulations. These were then re-categorised ( Figure 2 ) using literature analysis into opioid/alkaloid candidate analgesics (alkaloids related to known opioid receptor ligands or ion channel ligands, 4 convergent compounds) ( Bienz et al, 2002 ; Kai et al, 1998 ; National Center for Biotechnology Information ; Nichols and Lee, 2018 ; Qi et al, 2019 ; Saeed et al, 1997 ; Song et al, 2000 ; Xu et al, 2005 ; Zhong et al, 1995 ), potential ligands for nociceptive ion channels (terpenes, 49 convergent compounds), components with other demonstrated neuroactivity ( 15 convergent compounds), components with bioactivity indirectly related to pain (anti-inflammatory, anti-oxidants, 16 convergent compounds) and compounds with other types of bioactivity but no obvious link to analgesia (56 convergent compounds) ( Antwi et al, 2018 ; Bommareddy et al, 2019 ; Crupi and Cuzzocrea, 2017 ; Devi et al, 2015 ; Do et al, 2019 ; Henry and Maisch, 1890 ; Herout and Sykora, 1958 ; Huang et al, 2015 ; Hui et al, 2015 ; Kimura et al, 2001 ; Ko et al, 2017 ; Kumar and Muller, 2000 ; Kwon et al, 2018 ; Le et al, 2018 ; Li et al, 2009 ; Madeo et al, 2018 ; National Center for Biotechnology Information ; Ng et al, 2008 ; Qian, 2002 ; Shim et al, 2018 ; Wang et al, 2019 ; Ye et al, 2018 ; Zeisel, 1981 ). “…For instance, linoleic acid, existing in almost all the three herbs except for FJ and GC, exerts potent antioxidant, anti-inflammatory, metabolic regulation, anticancer, and immune-regulatory activities [38]. Similarly, quercetin, ursolic acid, stigmasterol, and angelicin, which were distributed in at least two herbs of KBL, also exert various pharmacological properties, participating in the regulation of multiple physiological and pathological processes, such as inflammation, immune response, and stress process [3942].…”
