Stimulation by ghrelin of p42/p44 mitogen‐activated protein kinase through the GHS‐R1a receptor: Role of G‐proteins and β‐arrestins

Abstract: Results presented in this study indicate that in human embryonic kidney 293 cells (HEK 293), the ghrelin receptor growth hormone secretagogue receptor type 1a (GHS-R1a) activates the extracellular signal-related kinases 1 and 2 (ERK 1/2) via three pathways. One pathway is mediated by the beta-arrestins 1 and 2, and requires entry of the receptor into a multiprotein complex with the beta-arrestins, Src, Raf-1, and ERK 1/2. A second pathway is G(q/11)-dependent and involves a Ca(2+)-dependent PKC (PKCalpha/beta)… Show more

“…It was not clear Rsk , Raf-1, PAK5, and 14-3-3 exist as complexes in some cell types (38,39,(47)(48)(49)(50). These complexes can regulate activity of some proteins acting through scaffold proteins (39,50,51). It is not yet known if B-Raf regulates NHE1 through part of a complex, although we did find that p90…”

“…However, like many other GPCRs (Gudermann et al, 1996;Wise et al, 1997), it is also able to modulate cellular signaling via pathways initiated via activation of other heterotrimeric G proteins (Holst et al, 2005;Camiñ a et al, 2007;Dezaki et al, 2007). When membranes of HEK293 cells transfected transiently to express both the ghrelin receptor and the G protein G␣ o1 were employed in […”

“…Src activation through beta-arrestin and GPCR has been reported outside of photoreceptors [49, 50]. Immunohistochemistry has shown the presence of beta arrestins in photoreceptors [51].…”
