Stimulation by leukotriene D4 of increases in the cytosolic concentration of calcium in dimethylsulfoxide-differentiated HL-60 cells.

Abstract: The C6-sulfidopeptide leukotrienes C4 (LTC4) and D4 (LTD4) evoked increases in the cytosolic concentration of intracellular calcium (QCa`2Ij) in dimethylsulfoxide-differentiated HL-60 cells, as assessed by the fluorescence of quin-2. The increases in 1Ca"2Ji reached a peak within 15-90 s, attained 50% of the maximum level at 1.2 nM LTD4 and 60 nM LTC4, were greater in maximal magnitude for LTD4 than LTC4, and sub-

“…These results are in keeping with previous results using HEK-293 cells expressing the CysLT 1 receptor [9] and human neutrophils [5], which showed that most of the response (>80%) persisted after removing extracellular calcium. Furthermore, in agreement with previous results with DMSO-differentiated HL-60 cells [32] and human neutrophils [5] [32,33], almost exclusive release from intracellular stores [5], or both [34].…”

“…In agreement with the results obtained with DMSOdifferentiated U937 [35] and HL-60 cells [32], LTD 4 caused homologous desensitization of the Ca 2+ mobilization in DSMCs. These data show that the desensitization produced by pretreatment with LTD 4 was selective and suggests that the CysLT 1 receptor is the receptor for LTD 4 , consistent with findings in other tissues [8,9].…”

“…LEUKOTRIENE RECEPTORS 561 through a PTX-sensitive G protein (G i/o ) (Baud et al, 1987). In further reports, investigators have shown that the Ca 2ϩ modulation may be through two distinct G proteins, one PTX-sensitive and one insensitive, in monocyte/macrophage U937 cells (Pollock and Creba, 1990;Capra et al, 2003) or in a human epithelial cell line (Sjölander et al, 1990;Adolfsson et al, 1996).…”

“…LTD 4 is known to induce either Ca 2ϩ influx through the plasma membrane without any Ca 2ϩ release from intracellular stores (Baud et al, 1987a;Oliva et al, 1994), or Ca 2ϩ release without influx (Bouchelouche et al, 1990) and in many cell types can induce both Ca 2ϩ influx and release (Mong et al, 1984b(Mong et al, , 1987a(Mong et al, , 1988Sjolander et al, 1990). In human airways, Gorenne et al (1998) -independent isoform of protein kinase C (PKC⑀).…”
