Stimulation of human peripheral blood lymphocytes by bioactive peptides derived from bovine milk proteins

Abstract: The in vitro modulation of the proliferation of human peripheral blood lymphocytes by different synthetic peptides derived from milk proteins was investigated. Therefore, proliferation changes were followed up after incorporation of BrdU into the DNA, and the influence on protein biosynthesis was measured using the [3H]leucine incorporation test. Tyr-Gly and Tyr-Gly-Gly significantly enhanced (maximal 90 and 35%, respectively) the proliferation of PBL. For 13-casomorphin-7 and 13-casokinin-10, lymphocyte proli… Show more

“…While cell proliferation is promoted by peptides from fish (Ravallec Ple et al 2001;Ravallec Ple et al 2000), soybean (Franek et al 2000), wheat (Franek et al 2000), and milk (Kayser and Meisel 1996), cell growth inhibition by peptides derived from soybean (Kops et al 1997) and milk (Ganjam et al 1997) has been described. Peptides derived from wheat have been found to inhibit (Giovannini et al 2000;Silano and De Vincenzi 1999) and promote (Rivabene et al 1999;Rocca et al 1983) cell death.…”

“…β-Casomorphin-7 derived from β-casein was used as a possible immunostimulating substrate, since it is a ligand of opioid receptors that have also been found on the surface of human T-lymphocytes (33)(34)(35).…”

“…Proliferation changes were measured as described in [17] using a colorimetric enzyme immunoassay (Boehringer Mannheim, Germany, kit 1647229) based on incorporation of 5-bromo-2'-deoxyuridine (BrdU) during DNA synthesis within 17 h (Caco-2) or 24 h (PBL, HL-60). Results were read out on an ELISA spectrophotometer (Anthos HT-2) at 450 nm (reference wavelength: 620 nm).…”
