Stimulation of in Vitro Granulocyte‐Macrophage Colony Formation by Mouse Heart Conditioned Medium

Abstract: Medium conditioned by mouse heart tissue was found to contain colony-stimulating factor(s) (CSF) which produced in vitro colonies of granulocytes and/or macrophages with bone marrow cells of CBA mice. The action of this CSF type (CSFMHCM) was compared with that of post-endotoxin mouse serum (CSFES on colony formation by density subpopulations of colony-forming cells (CFU-c), separated by isopycnic centrifugation in a continous albumin density gradient. The number, size and morphology of colonies were examined.… Show more

“…CFU-C were assayed according to the method of Bradley & Metcalf (1966) as modified by Iscove (1972). MNC 105 were cultured in 1 ml a-medium containing 0.8% methylcellulose, 20% horse serum and 20% mouse heartconditioned medium (Byrne et at., 1978).…”

“…This indicates that this MGI-1M differed antigenically from MGI-1G and MGI-2. The functional specificities of MGI-1M and MGI-1G may be associated with a preferential activity on specific normal myeloid progenitors in the heterogeneous progenitor population in the bone marrow (Haskill et al, 1970;Byrne et al, 1978).…”

“…Cells at a concentration of 0.2 x 1Q6Mml1 were cultured in a-medium containing 5% v/v newborn calf serum and 15% donor horse serum (Gibco). Mouse heart conditioned medium was used as a source of colony stimulating factor (Byrne et al, 1978).…”
