Stimulation of NFκB Activity by Multiple Signaling Pathways Requires PAK1

Abstract: The p21-activated kinase (PAK1) is a serine-threonine protein kinase that is activated by binding to the Rho family small G proteins Rac and Cdc42hs. Both Rac and Cdc42hs have been shown to regulate the activity of the transcription factor NFB. Here we show that expression of active Ras, Raf-1, or Rac1 in fibroblasts stimulates NFB in a PAK1-dependent manner and that expression of active PAK1 can stimulate NFB on its own. Similarly, in macrophages activation of NFB as well as transcription from the tumor necro… Show more

“…Either of these activities conceivably couples to regulation of NF-B and cyclin D1. For example, Rac can regulate NF-B and cyclin D1 induction through activation of p21-activated protein kinase and other downstream effectors (17,24,46). In contrast to negative regulation of Rac, sequestration of RalBP1 by Ral-GTP possibly elevates basal levels of active Rac.…”

“…Other groups have observed RelA/p65 nuclear translocation independent of the classical IKK-I B pathway: coexpression of PAK1 was shown to induce RelA/p65 nuclear translocation independent of IKK␣/IKK␤ activity or I B␣ Ser 32 /Ser 36 phosphorylation (53). Recently, p53 was shown to induce NF-B activation through RSK-1-mediated phosphorylation of RelA/p65 in an I B␣ degradation-independent manner (54).…”
