Methylphenidate (MPD), commonly known as Ritalin, is the most frequently prescribed drug to treat children and adults with attention deficit hyperactivity disorder (ADHD). Adolescence is a period of development involving numerous neuroplasticities throughout the central nervous system (CNS). Exposure to a psychostimulant such as MPD during this crucial period of neurodevelopment may cause transient or permanent changes in the CNS. Genetic variability may also influence these differences. Thus, the objective of the present study was to determine whether acute and chronic administration of MPD (0.6, 2.5, or 10.0 mg/kg, i.p.) elicit effects among adolescent WKY, SHR, and SD rats and to compare whether there were strain differences. An automated, computerized, open-field activity monitoring system was used to study the dose response characteristics of acute and repeated MPD administration throughout the 11-day experimental protocol. Results showed that all three adolescent rat groups exhibited dose-response characteristics following acute and chronic MPD administration, as well as strain differences. These strain differences depended on the MPD dose and locomotor index. Chronic treatment of MPD in these animals did not elicit behavioral sensitization, a phenomenon described in adult rats that is characterized by the progressive augmentation of the locomotor response to repeated administration of the drug. These results suggest that the animal's age at time of drug treatment and strain/genetic variability play a crucial role in the acute and chronic effect of MPD and in the development of behavioral sensitization.

INTRODUCTIONSubstance dependence is one of the most established physiological states following chronic exposure to an addictive drug. It is characterized by a cluster of physio-behavioral and cognitive phenomena which include the persistent desire or unsuccessful efforts to cut down substance abuse, unrestrained drug taking despite harmful consequences, higher priority given to drug use than to other social, recreational activities and obligations, tolerance to the drug's euphorigenic effects and a physical withdrawal state (International Classification of Diseases (ICD)-10).Measuring the dependence liability of a certain substance is a challenging task as some components of dependence cannot be gauged objectively in the laboratory. Pro-addictive behaviors such as tolerance and withdrawal can be well studied in animal subjects and they have provided us with important information on the effects of drugs on health and longterm consequences of substance use. However, it is diffi cult to measure the cognitive aspect of dependence (such as drug craving, loss of control and persistent drug use), although this is slowly being addressed by recent neuroimaging techniques Repeated administration of addictive drugs causes cellular and molecular changes believed to be the mechanism of pro-addictive behaviors. Neuroadaptations also take place with repeated administration of amphetamine, methylphenidate and atomoxetine, drugs for Attention Defi cit Hyperactivity Disorders (ADHD), and it is speculated that these changes may serve as markers to demonstrate the dependence liability of these therapies. In this review, we enumerate the neuroadaptive changes in molecules associated with neuronal signaling and plasticity, as well as neuronal morphology wrought by repeated administration of ADHD medications. We provide the current perspective on the dependence liability of these therapies, and also suggest of some factors that need to be considered in future investigations, so that what is drawn from animal studies would be better consolidated with those known clinically. Objective: The present study aimed to evaluate the neuro-protective and hepato-protective activity of melatonin (MLN) on non-alcoholic steatohepatitis (NASH) in albino rats induced by methionine and choline deficient (MCD) diet. Material and Methods: To achieve this aim, rats were fed MCD diet for 8 weeks to induce non-alcoholic steatohepatitis. Oral administration of MLN (10 mg/kg/day) was done for the last two consecutive weeks. Twenty-four hours after the last dose of the MLN behavioral test, namely activity cage test was performed and the activity counts were recorded. Serum AST, ALT, ALP, GGT, total/direct bilirubin and albumin were valued to assess liver function. Moreover, hepatic interleukin-6 (IL-6) as well as nuclear factor-kβ (NF-kβ) were determined. Besides, brain biomarkers viz. ammonia, nitric oxide and brain-derived neurotrophic factor (BDNF) were measured which are reliable indices to assess brain damage. Results: MLN succeeded in suppression of NASH rats' locomotor activity. Besides; it improved all biochemical parameters and histopathological as well as proliferating cell nuclear antigen (PCNA) immunohistochemical profiles of liver and brain. Conclusions: These results afford new intuition into the possible molecular mechanisms by which MLN faces steatohepatitis disorder, via modulation of several factors participating in the progression of this disease.
