Strain-Specific Gifsy-1 Prophage Genes Are Determinants for Expression of the RNA Repair Operon during the SOS Response in Salmonella enterica Serovar Typhimurium

Abstract: The transcriptional activator RtcR and the RNA repair proteins whose expression it regulates, RtcA and RtcB, are widely conserved in Proteobacteria . In Salmonella Typhimurium 14028s, genotoxic stress activates RtcR to direct RpoN-dependent expression of the rsr-yrlBA-rtcBA operon.

“…Finally, three translocated proteins, CbeA, YagB, and YfJZ, are antitoxins characteristically encoded within prophages ( Table 1 ). This finding is of interest given that RecA and the SOS response are known inducers of prophage lytic programs and toxin-antitoxin systems ( 81 , 82 ). Conceivably, translocation of these antitoxins serves to block phage-encoded or other toxins produced as a component of the mating-induced SOS response.…”

“…1F). As it appears that oxidative DNA damage activates prophage simultaneously with RNA repair ( 15 ), we investigated whether Gifsy prophage activation caused tRNA cleavage. A strain of S. enterica lacking all three Gifsy prophages failed to induce tRNA Leu cleavage after exposure to H 2 O 2 (Fig.…”

“…The data so far indicated the presence of a previously uncharacterized nuclease in the Gifsy-1 prophage that can target its bacterial host’s tRNA molecules. We initially focused on the STM14_3218-20 operon within the Gifsy-1 prophage, a locus that is associated with the activation of rtc gene transcription after antibiotic-mediated DNA damage ( 15 ) and that is present in Salmonella Typhimurium strain 14028s but absent in S. Typhimurium strains LT2 or SL1344. Northern blot analyses showed that H 2 O 2 induced tRNA Leu cleavage in S. Typhimurium strains LT2, SL1344—as well as in S. Typhimurium 14028s mutants bearing multiple- or single-gene deletions in the STM14_3218-20 operon—as efficiently as in the WT 14028s strain (fig.…”

“…Furthermore, the identification of the prophages Gifsy-1, Gifsy-2, Fels-1, and Fels-2 in S. Typhimurium has been linked to an adaptative response to stress conditions. 50 Notably, studies have established that the Gifsy prophage family may genetically contribute to the presence of the…”
