Strategies for Natural Product Discovery by Unlocking Cryptic Biosynthetic Gene Clusters in Fungi

Abstract: Recent advancements in genome sequencing and bioinformatic analysis of fungal genomes have revealed that fungi possess cryptic or silent biosynthetic gene clusters (BGCs). This discovery suggests that our understanding of the universe of fungal secondary metabolomes is limited. In this review, we summarize recent strategies for activating cryptic BGCs in fungi, identify fungal secondary metabolites, and highlight their biological activities. We also briefly discuss the isolation and purification methods for th… Show more

“…Even with the immense genetic similarity, not all genes and gene clusters evaluated are expressive, and they may be inactive (Onaka 2017;Hur et al 2023). In total, 882 putative proteins identified through protein prediction tools indicated the presence of new gene clusters, some of which remain dormant or unexpressed.…”

“…This is an unusual sesquiterpene alcohol synthesized by a sesquiterpene cyclase from Trichoderma sp. [70]. In region 95.1, one NRPS was found to be accountable for the biosynthesis of ilicicolin H. This compound is a broad-spectrum antifungal agent that acts…”

“…In many cases, these limitations arise from the difficulty in isolating pure active substances from endophytes and the low yields of the resultant compounds—issues that are further complicated by the fact that some of the compounds are synthesized only during a limited phase of the endophytes’ life cycle [ 28 , 29 , 30 ]. Therefore, it is important to identify biosynthetic gene clusters for biologically active secondary metabolites of endophytes [ 31 ]. With such identification, it would be possible to dissect the biosynthesis pathway of a given secondary metabolite and the main genes encoding the enzymes responsible for the biosynthesis.…”
