Stress, Genes and the Biology of Suicidal Behavior

Abstract: Suicidal behavior is in part heritable. Studies seeking the responsible candidate genes have examined genes involved in neurotransmitter systems demonstrated to have altered function in suicide and attempted suicide. These neurotransmitter systems include the serotonergic, noradrenergic and dopaminergic systems and the HPA axis. With some exceptions, most notably the serotonin transporter promotor HTTLPR polymorphism, replication of candidate gene association studies findings has proven difficult. This chapter… Show more

“…Thus, sibling analysis should account for genetic confounding. A hereditary component has been suggested for suicidal acts [3] and smoking [25], and the search for candidate genes has recently started for both suicidal [26][27][28] and smoking [25] behaviors. Sibling analysis also control for important environmental factors shared by the siblings.…”

“…Вероятность завершенного суицида у монозиготных близ-нецов примерно в 20 раз выше, чем у дизиготных, но у дизиготных близ-нецов -в 3,5 раза выше среднего уровня в популяции. У приемных детей, в семейном анамнезе которых были сведения о суициде, частота суици-дальных попыток в 6 раз выше по сравнению с контрольными приемными детьми, чьи биологические родители не совершали таких попыток (Currier et al, 2008).…”

“…Although the underlying mechanism between adverse life events and suicide remains unclear, according to the stress-diathesis model, adverse life event(s) can trigger a series of genetic, physiological, and psychological changes including the modification of gene expression through DNA methylation [1,53,[68][69][70] . A longitudinal prospective study showing that depressive mood was a necessary precondition of suicide ideation after adverse events occurred [71] .…”
