Structural and In Vitro Functional Comparability Analysis of Altebrel™, a Proposed Etanercept Biosimilar: Focus on Primary Sequence and Glycosylation

Abstract: The demand for reliable comparability studies of biosimilars grows with their increased market share. These studies focus on physicochemical, structural, functional and clinical properties to ensure that a biosimilar has no significant differences to the originator product and can be released into the market without extensive clinical trials. In the current study, Enbrel® (etanercept, the originator) and Altebrel™ (the proposed biosimilar) underwent direct comparison. “Bottom-up” mass spectrometric analysis wa… Show more

“…Lately, deconvolution of CD spectra ( Fazel et al, 2019 ; Hermosilla et al, 2020 ) and IR absorption spectra (1700–1,600 cm −1 ) ( Lee et al, 2019 ; Shekhawat et al, 2019 ) using different software (K2D2, BeStSel, OriginPro, etc.) have been successfully used to quantitate secondary conformations (i.e., alpha-helix, beta-sheet, or random coil).…”

“…Samples were injected by loop overfilling into a 5μL loop. For the 60 min [ 33 ] LC gradient, the mobile phase consisted of solvent A (0.2% v/v formic acid and 3% DMSO v/v in water) and solvent B (0.2% v/v formic acid and 3% DMSO in EtOH) at a flow rate of 3 μl/ min [ 32 , 33 ].…”
