Phaseolus vulgaris (common bean) can be nodulated by several Rhizobium species. Among them, Rhizobium tropici has a relatively broad host range, as it is able to infect beans, Leucaena trees, and several other legumes. This work describes the isolation and the characterization of extracellular factors (Nod factors) whose production from R. tropici was triggered by the transcriptional activation of its nod genes. These factors consist of a chitopentaose backbone in which the N-acetyl group of the nonreducing end glucosaminyl residue is replaced by an N-methyl-N-vaccenoyl one. Some of these molecules are sulfated on position 6 of the terminal reducing glucosamine.

Lipo-chitooligosaccharides (LCOs) were originally found as symbiotic signals called Nod Factors (Nod-LCOs) controlling nodulation of legumes by rhizobia. More recently LCOs were also found in symbiotic fungi and, more surprisingly, very widely in the kingdom fungi including in saprophytic and pathogenic fungi. The LCO-V(C18:1, Fuc/MeFuc), hereafter called Fung-LCOs, are the LCO structures most commonly found in fungi. This raises the question of how legume plants, such as Medicago truncatula, can discriminate between Nod-LCOs and these Fung-LCOs. To address this question, we performed a Genome Wide Association Study on 173 natural accessions of Medicago truncatula, using a root branching phenotype and a newly developed local score approach. Both Nod- and Fung-LCOs stimulated root branching in most accessions but the root responses to these two types of LCO molecules were not correlated. Also, heritability of root response was higher for Nod-LCOs than for Fung-LCOs. We identified 123 loci for Nod-LCO and 71 for Fung-LCO responses, but only one was common. This suggests that Nod- and Fung-LCOs both control root branching but use different molecular mechanisms. The tighter genetic constraint of the root response to Fung-LCOs possibly reflects the ancestral origin of the biological activity of these molecules.

Rhizobium lipochitooligosaccharide signal molecules stimulate multiple responses in legume host plants, including changes in host gene expression, cell growth, and mitoses leading to root nodule development. The basis for signal transduction in the plant is not known. We examined cytoplasmic free calcium in host root hairs using calcium-sensitive reporter dyes. Image analysis of injected dyes revealed localized periodic spikes in cytoplasmic calcium levels that ensued after a characteristic lag following signal application. Structural features of the signal molecules required to cause nodulation responses in alfalfa are also essential for stimulating calcium spiking. A nonnodulating alfalfa mutant is defective in calcium spiking, consistent with the possibility that this mutant is blocked in an early stage of nodulation signal perception.
