Structural mass spectrometry analysis of lipid changes in a Drosophila epilepsy model brain

Abstract: Phosphatidylethanolamine (PtdEtn) is one of the most abundant phospholipids in many animal cell types. The Drosophila easily shocked (eas2) mutant, used as an epilepsy model, is null for the PtdEtn biosynthetic enzyme, ethanolamine kinase. This mutant displays bang sensitive paralysis, and was previously shown to have decreased levels of PtdEtn. We have developed a highly selective and sensitive measurement strategy using ion mobility-mass spectrometry for the relative quantitation of intact phospholipid speci… Show more

“…Recently IM-MS measurement strategies have demonstrated great utility in the analysis of highly complex biological samples ranging from serum [51] to intact tissue [24; 75; 76]. The advantages of IM-MS for complex sample analysis are several fold, including (i) simultaneous characterization of all biomolecular species in a sample, (ii) minimizing sample losses and sample preparation artifacts arising from extensive purification procedures, (iii) preserving the biomolecular context, such as that encountered with conjugate species ( e.g.…”

“…Recently, IM-MS was utilized by Kliman et al to separate lipid signals from matrix cluster signals in a relative quantitation study of lipids from a Drosophila eas 2 epilepsy model mutant. By removing the extraneous contribution of matrix cluster signals from the lipid analysis, statistically significant changes in the abundance of several lipid species were observed between mutant and wildtype brain tissue samples [76]. …”

“…CDP-ethanolamine donates phosphoethanolamine to diacylglycerol to generate PE, which is mediated by CDP-ethanolamine phosphotransferase (encoded by bbc ) (Dobrosotskaya et al, 2002). In addition, the level of PE is reduced in eas whole flies and heads (Kliman et al, 2010; Nyako et al, 2001; Pavlidis et al, 1994). Therefore, we asked whether the dendrite morphogenesis defects were due to insufficient PE synthesis through the CDP–ethanolamine pathway.…”
