Structure-Activity Relationships of Saponins fromGleditsia sinensisin Cytotoxicity and Induction of Apoptosis

Abstract: Thirteen compounds were isolated from the anomalous fruits of Gleditsia sinensis on the basis of bioassay-guided fractionation. These saponins together with six analogues or related compounds were tested for their cytotoxicities against six tumor cell lines by the MTT method. The induction of apoptosis in HL-60 cells by these compounds was determined through flow cytometric analysis. Some structure-activity relationships in cytotoxicity and induction of apoptosis were identified. The evaluation of the cytotoxi… Show more

“…According to the literature, the effects of saponins on the digestive system are diverse because they can act both as protectors against ulcerative lesions and as destructors of the cellular membrane. Saponins have also been described as enhancers of gastric permeability and natural enhancers of tumor cell apoptosis (Chow et al, 2002;Zhong et al, 2004), supporting the notion that the biological effects of saponins vary among species.…”

“…Chinese herbal antitumor agents, including assolanine (6), an extract from Rhodiola rosea rhizomes (7), Gleditsioside E from Gleditsia sinensis (8) and saponins from Gleditsia sinensis (9), exert significant effects against a variety of tumors and have been confirmed to be effective antitumor agents. Saffron is a derived plant product from the dried stigma of the Crocus sativus flower (family, Iridaceae), which has been used for several hundred years in the treatment of cardiovascular diseases, inflammatory diseases and several other tumors (10,11).…”

“…Twelve pure saponins were isolated from G. sinensis, and were fully characterized as reported in our previous paper [9]. They include gleditsia saponins B, C, and C′ (abbreviated as GSB, GSC, and GSC'), and gleditsiosides A-D, F, G, N, O, and Q (abbreviated as GA, GB, GC, GD, GF, GG, GN, GO, and GQ).…”

“…Moreover, they could be acylated by one or two monoterpenic acid groups at the α-chain. The monoterpenic acid residue, either 6(R)-6-hydroxy-2,6-dimethyl 2,7-octadienolic acid (menthiafolic acid) or 6(R)-6-hydroxy-2-hydroxymethyl-6-methyl 2,7-octadienolic acid, is critical for the cytotoxic activities of these saponins [9]. Although Gleditsia saponins appear to be a promising antitumor agent, the composition has not been well addressed thus far.…”
