Structure and Properties of Tethered Bilayer Lipid Membranes with Unsaturated Anchor Molecules

Abstract: The self-assembled monolayers (SAMs) of the new lipidic anchor molecule HC18 [Z 20-(Z-octadec-9-enyloxy)-3,6,9,12,15,18,22-heptaoxatetracont-31-ene-1-thiol], and mixed HC18/β-mercaptoethanol (βME) SAMs were studied by spectroscopic ellipsometry, contact angle measurements, reflection adsorption infrared spectroscopy (RAIRS), electrochemical impedance spectroscopy (EIS), and evaluated in tethered bilayer lipid membranes (tBLMs). Our data indicate that HC18, containing a double bond in the alkyl segments, forms … Show more

“…The plasmid used for the ϩmyrMA preparation via coexpression of (31). Lipid mixtures at the desired molar ratios were prepared from fresh lipid stock in chloroform, and the solvent was evaporated under vacuum for 12 h. The dried lipid films were hydrated in a high-salt aqueous buffer (1 M NaCl, 10 mM NaPO 4 [pH 7.4]) to a lipid concentration of ϳ5 mg/ml and subsequently sonicated until clear lipid solutions were obtained.…”

“…1) incorporates a single lipidic bilayer membrane of wellcontrolled composition that is tethered to a planar electrode-a 0.05-m-thick Au film on a glass or Si substrate-via short ethyleneoxide anchors (31), thereby providing a hydrated environment on both sides of the membrane. Thereby, the bilayer retains its intrinsic fluidity (32), is virtually defect-free (29), and is resilient to external manipulations such as buffer exchanges, as monitored by electrochemical impedance spectroscopy (33).…”

“…Both the SPR method for quantifying intermolecular interactions (51) and the sparsely tethered bilayer lipid membrane (stBLM) model membrane system (52) are well established. stBLMs incorporate a single lipidic bilayer membrane of controlled composition that is tethered to a planar electrode-a 0.05-m-thick Au film on a glass or Si substrate-via short ethylene oxide anchors (53), thereby providing a hydrated environment on both sides of the membrane. Thus, the bilayer retains its intrinsic fluidity (54), is virtually defect free (52), and is resilient to external manipulations, such as buffer exchanges, as monitored by electrochemical impedance spectroscopy (EIS) (55).…”

“…The tether lipid for supported membranes, Z 20-(Z-octadec-9-enyloxy)-3,6,9,12,15,18,22-heptaoxatetracont-31-ene-1-thiol (HC18) (17), was a generous gift from Dr. David Vanderah (Institute for Bioscience and Biotechnology Research, Rockville, MD). ␤-Mercaptoethanol was from Sigma-Aldrich.…”
