Structure, expression, and regulation of the murine renin genes.

Abstract: It has long been known that the renin-angiotensin system plays an integral role in the regulation of blood pressure and electrolyte and fluid balance in mammals. The advent of molecular biologic techniques has afforded new insights into the genes regulating blood pressure. Laboratory mice and rats have been used as experimental models to examine the structural organization and expression of the renin gene. It is now well established that some mice, unlike rats and humans, contain a duplicated copy of the renin… Show more

“…Although renin and renin mRNA may be expressed in many extrarenal cells, such as the placental, ovarian, testicular, 35 adrenal, 36 and pituitary 37,38 cells of a number of species, the juxtaglomerular cells of the kidney are thought to be the only source of circulating renin. 39 This explains the parallel changes of plasma renin activity and kidney renin levels elicited by chronic Ang II infusion and chronic ARB treatment.…”

“…Ren-1 and Ren-2 share 97% amino acid identity (3). The Ren-1 protein is the major source of circulating renin and thus believed to be the major regulator of the reninangiotensin cascade.…”

“…Our previous studies have demonstrated that 1,25-dihydroxyvitamin D 3 (1,25(OH) 2 D 3 ), the hormonal form of vitamin D, is a crucial negative regulator of renin biosynthesis (24). The repressive activity of 1,25(OH) 2 D 3 is mediated by the vitamin D receptor (VDR), a member of the nuclear receptor superfamily (25).…”

“…Most mouse models have been generated by gene targeting using embryonic stem (ES) cells of strain 129 (2). This strain differs from humans in having two renin genes in tandem on the same chromosome (Ren2 and Ren1d) that differ in their expression and regulation in ways that complicate interpreting experimental data (3). For example, Ren2 can compensate for the effects of the absence of Ren1d on BP and kidney development (4).…”
