“…The absence of kpsU is consistent with the lack of elevated levels of CMP-KDO synthetase found in group III-expressing strains (18). The identification of homologues to the KpsC and KpsS proteins is in keeping with the observation that phosphatidyl-KDO has been detected at the reducing end of the K10 polysaccharide (39) and would be in agreement with the suggested role of the KpsC and KpsS proteins in the addition of phosphatidyl-KDO to the reducing end of polysaccharide (34). The lack of a capsulespecific CMP-KDO synthetase enzyme would suggest that the CMP-KDO for group III capsule expression is supplied by the KdsB enzyme (11,28).…”

“…The rmlC gene encodes dTDP-4keto-6-deoxy-D-glucose-3,5-epimerase, which is required for synthesis of an intermediate in the dTDP-rhamnose pathway (47). The E. coli K10 capsule contains rhamnose as one of its component sugars (39), and so this RmlC homologue is probably involved in the synthesis of dTDP-rhamnose required for biosynthesis of the K10 polymer. Therefore, rmlC K10 defines the boundary between the serotype-specific region 2 and a conserved region 3 involved in capsule export.…”

“…Second, antibody and bacteriophage specific for the K10 capsule were available, facilitating isolation of K10-expressing clones. Finally, the structure of the K10 capsule was known (36). A cosmid library of E. coli D1114 chromosomal DNA was constructed in E. coli LE392 and screened by colony immunoblotting with polyclonal antiserum to the K10 capsular polysaccharide.…”
