Studies on Anti-Depressant Activity of Four Flavonoids Isolated from Apocynum venetum Linn (Apocynaceae) Leaf in Mice

Abstract: Purpose: To investigate the anti-depressant activity of kaempferol, quercetin, kaempferol-3-O-β-Dglucose and quercetin-3-O-β-D-glucose isolated from Apocynum venetum (72.58 -90.24; 52.58 -70.24 s), 5-HIAA levels (940.8 -1244.7; 880.8 -1164.1 ng/g) and 5-HIAA/5-HT ratio (1.77 -4.76; 1.83 -4.16), but increased NE, DA and 5-HT levels (238.7 -405.7, 308.4 -528.1, 261.4 -531.9; 243.9 -423.6, 296.7 -534.9, 279.8 -481.4 ng/g) in FST and TST, compared with control group (146.18, 126.18 s; 1363.4, 1240 7.43, 6.16; 1… Show more

“…Among these compounds, flavonols are the major flavonoids. This result is consistent with the previous data [20,21], while more anthocyanins were detected. Quantitative analysis shows that flavonols are the main differentiated flavonoids between the A. venetum and A. hendersonii leaf samples.…”

“…Yan et al reported that kaempferol and quercetin have potential antidepressant-like activity. The mechanism was attributed to the increased levels of NE, dopamine (DA) and 5-HT [ 33 ]. Another study on quercetin proposes its antidepressant-like activity through its inhibitory effect on MAO-A, which is an important enzyme in the metabolism of the neurotransmitter, 5-HT.…”

“…Quercetrin (tetrahydroxyflavone) is a quercetin O-glycoside, in which quercetin is substituted with α-L-rhamnosyl moiety at position C3 via glycosidic linkage ( Figure 22 ) [ 100 ]. In another study, kaempferol, kaempferol-3-O-β-D-glucoside, quercetin, and quercetin3-O-β-D-glucoside ( Figure 23 ) isolated from hydroethanolic extract of Apocynum venetum L. leaves, were also found to exert significant anti-depressant actions in mice [ 101 ]. The underlying mechanism behind this effect was suggested to be increase in dopamine, serotonin and nor-epinephrine level with reduction in serotonin metabolism [ 101 ].…”

Kaempferol-3-O-β-D-glucoside is a trihydroxyflavone, which is also known as kaempferol O-glucoside and having glucosyl residue at position C3 of kaempferol via a β-glycosidic linkage ( Figure 24 ).…”
