Studies on structure-function relationships of acetolactate decarboxylase from Enterobacter cloacae

Abstract: Arg150 in E.c.-ALDC exhibited a unique tilted conformation implying the lower activities of E.c.-ALDC comparing to other bacterial ALDCs. Sequence conservation of E.c.-ALDC is represented by ribbons and lines (conserved, magenta to variable, cyan).

“…Based on that knowledge, new biotechnological products may be developed and innovative solutions may be introduced that exploit plant-beneficial bacteria for biological control of plant diseases (biopesticides) and for plant growth promotion (biofertilizers) for sustainable agricultural practices and phytoremediation (Mesa-Marín et al, 2020). “…Of these two enzymes, only kinetic parameters for ALDC from E. cloacae ssp. dissolvens are available: the K M is 12.2 mM, and k cat is 0.96 s −1 [ 47 ] while known ALS enzymes have a turnover number 100 times greater [ 48 ]. Even without kinetic parameters for the specific ALS used, the low affinity turnover rate of ALDC, taken together with the observed accumulation of acetolactate, support the hypothesis that ALDC is the rate limiting step for acetoin formation.…”
