Mast cells were demonstrated in eight species of snakes, using special fixation techniques to prevent solubilization of cytoplasmic granules. Toluidine blue 0 and azure A were the major stains and observations were made under light microscope including cytophotometric analysis.The mast cells of snakes were shown to be relatively small (7-11 p in diameter) when compared to mast cells of a lizard (8-15 p), dog and rat (9-15 p).Among the various organs examined, mast cells were particularly numerous in the mesentery, tongue, underneath the serosa of the digestive tract and in the heart, between muscle fibers and i n the epicardium.Although under the light microscope some snake mast cells seemed to be orthochromatically stained, when analysed by cytophotometry they were demonstrated to be actually metachromatic.Snake mast cells granules were demonstrated to have an amphoteric behavior, since they were stained with both basic (toluidine blue 0 and azure A) and acid dyes (eosin and ponceau-acid-fuchsin). 109

In the ultrastructural comparison of normal, unimmunized spleens with immunized spleens at key intervals after antigenic stimulation with keyhole limpet hemocyanin (KLH), we noted cellular and cytological features which reflect the cellular kinetics of the primary immune response, particularly with respect to plasma cell production. Although lymphoblasts and mature plasma cells are present in the white and red pulp, respectively, intermediate stages of the plasma cell line are rarely found in normal spleen. Following antigenic challenge, we found a marked increase in lymphoblasts in the white pulp, most of them containing short segments of rough endoplasmic reticulum suggesting initial differentiation toward plasma cells. Following an apparent migration of cells from the white to the red pulp, we found plasma cells in various stages of maturation in the red pulp cords and sinuses. The ultrastructural features of these cells reflect the differentiation of lymphoblasts into mature plasma cells. Both immature and mature plasma cells usually possess dilated cisternae of rough endoplasmic reticulum, suggesting that they are capable of producing and storing a secretory product, presumably antibody. We also noted a large number of immature macrophages and monocytes in immunized spleens. These cellular events and their cytological characteristics are compared to those described in other vertebrate classes.

The amphisbaenians, to judge from the many changes of opinion regarding their appropriate position among the vertebrates, appear to have puzzled most students of herpetology. Re‐evaluation suggests that a case may now be made for classifying the amphisbaenians as an order, separate from the lizards and snakes, but included with them in a superorder Squamata. Evidence for such a position may be derived from several dozen systems in which amphisbaenians differ from lizards, from snakes, or from both. Although individual character states are occasionally equivalent to those shown by one or another family of lizards, there is no “closest” family. This suggests that the Amphisbaenia probably diverged from the remainder of the Squamata before the diversification of all present families of lizards. Unfortunately many of the character states are still known from only a limited number of amphisbaenians and information for other groupings of squamates is similarly inadequate.
