Sickle cell disease (SCD) is a common recessively inherited disorder of hemoglobin affecting peoples everywhere in the world. The homozygous state (SS) is associated with complications and reduced life anticipation. The symptoms including, shortness of breath, heart palpitations, abdominal pains, aches and pains in the muscle.One cause of this disease is oxidative stress which is an imbalance between the reactive oxygen species and the potential of the body to neutralize these reactive species. An anti-oxidant is a substance which prevents oxidative stress by scavenging the free radicals. Medicinal plants are good and rich sources of natural antioxidants. Crude extracts from plants have been used in treating many diseases since ancient times. Various advances in scientific research on the use of plants and herbs explore the beneficial aspects of traditional medicine. This review highlights the role of oxidative stress in the progression and development of Sickle Cell Disease (SCD) and reviews the available literature reporting antisickling properties of a number of plants extracts.

The in vitro effects of aqueous extracts of Zanthoxylum macrophylla roots on the activities of three ATPases (Na + , K + -; Ca 2+ -; and Mg 2+ -ATPases) were investigated in membrane preparations from human erythrocytes of HbAA, HbAS and HbSS bloods. The activities of these membrane-bound enzymes were determined in the presence of varying concentrations of the plant extract. The results indicated that for Na + , K + -and Ca 2+ -ATPases, a dose-dependent activation of the three enzymes by the aqueous plant extract occurred. The trend for the different genotypes was HbSS>HbAS>HbAA. The activations were significant (p<0.05) when compared with activities obtained in the absence of the plant extract. Phenylalanine, a known anti-sickling agent, at varying concentrations (200-800µM) produced similar activating effect. In contrast, the extract inhibited the Mg 2+ -ATPase activity in a dose-dependent manner with a trend of HbSS>HbAS>HbAA. The inhibitions seen were significant (p<0.05) in comparison with activities in the absence of the plant extract. Phenylalanine produced similar effect for Mg 2+ -ATPase. These findings in this study suggest that differences in the ATPases of the different genotypes may account for the reports of other workers on the involvement of ion transport as a factor in the physiological manifestation of sickle cell disease. The findings are also significant to an understanding of the molecular basis for the use of the aqueous extract of Z. macrophylla roots in the management of sickle cell disease.

Antisickling activities of extracts of leaf, seed and seed pod of Garcinia kola Heckel

Garcinia kola has been hitherto speculated as effective in the management of sickle cell disease (SCD). Investigations into the antisickling activities of both crude methanol extracts and the aqueous fractions of the leaf, seed and seed pod of G. kola were carried out using p-hydroxybenzoic acid and normal saline as positive and negative controls respectively. At the tested concentrations of 10.0, 1.0 and 0.1 mg/ml, the leaf extracts exhibited greatest antisickling activity whilst the seed pod had the least antisickling activity. However, it was observed that the activity of methanol extract of the seed pod did not differ significantly (p>0.05) from that of the positive control, p-hydroxybenzoic acid. Results for screened phytochemicals of the investigated parts revealed the presence of saponins, tannins, combined and free anthraquinones. The results infer a preliminary confirmation for the effectiveness and use of Garcinia kola in the management of SCD and its implication in drug development.
