Studies on the Uronic Acid-Containing Glycoproteins of Fusarium sp. M7-1: II. The Primary Structures of the Low Molecular Weight Carbohydrate Chains of the Glycoproteins

Abstract: The primary structures of the O-glycosidically linked oligosaccharides isolated from glycoproteins GP I and GP II of Fusarium sp. M7-1 were established. The oligosaccharides released by alkaline borohydride treatment from the glycoproteins were purified by Bio-Gel P-4 and HPLC. This approach resulted in one monosaccharide and seven oligosaccharides. Their primary structures were resolved mainly by NMR spectrometry in combination with methylation mass spectrometry and fast atom bombardment mass spectrometry. Th… Show more

“…Siddiqui and Adams were first to report the presence of an extracellular galactofuranosecontaining glycoprotein in Gibberella fujikuroi (Fusarium moniliforme) [20]; only few structural elements were then presented. To our knowledge, mycelium glycoproteins of only one Fusarium sp., designated M7-1, were elucidated in details [21][22][23]. Fusarium sp.…”

“…Other minor oligosaccharide chains, released from the glycoprotein by mild alkaline treatment, were also characterized. They were composed mainly of Man residues with α-(1,2)-linkages but also contained GlcNAc, Rha, Man-6-phosphate [24], Man-6-phosphoethanolamine [21], and Man-6-phosphocholine [25]. The same authors described the production of extracellular acidic glycoproteins by several Fusarium species, and characterized the structure of a glycoprotein from F. oxysporum.…”

“…the signals other than those of choline are identical \'lith those of Maned -2Man-ol. 2) In the NOE difference spectrum of the isolated compound (data not shown), NOE was observed at the C-2 proton of mannitol with irradiation at the H-I signal of the mannose residue, indicating that the mannose residue is linked to the C-2 of mannitol. The data of the chemical shift (5.01 ppm) and J value (2.0 Hz) of the H-I signal of mannose residue and NOE difference spectrum indicated that mannose is linked to C-2 of mannitol through añ -linkage.…”

“…We demonstrated 2 ) that the primary structures of low molecular weight carbohydrate chains of the glycoproteins e( of Fusarium sp. M7-1 are composed mainly of mannose residues with mannose or mannose-6-phosphate ethanolamine as the reducing terminal, and that the mannose residues are attached to serine and/or thereonine through an O-glycosidic linkage.…”
