Carbon nanohorns (CNHs) are attractive for various applications, where a high specific surface area and long dispersion stability in water are important. In the present work, we study these parameters of CNHs prepared by arc evaporation of graphite depending on the conditions of the synthesis and subsequent oxidation in air. It is shown that the addition of toluene in the reactor during the arcing allows obtaining CNHs functionalized with −CHx groups. Heating of CNHs in air at 400 °C leads to substitution of −CHx groups for oxygen-containing groups. Moreover, the CNH endcaps are opened at 500 °C, and as a result, the specific surface area of CNHs increases 4 times. Aqueous suspensions with a concentration of oxidized CNHs of 100 µg/mL are stable for 8 months.

Fullerenes have become very popular nowadays in the field of cosmetic and medicine as “radical sponge” because of their high antioxidant activities. However, there are worries about the toxicity of fullerenes due to their special chemical properties and good ability to enter the human body. In this article, it is found that fullerenes have been proved to have the ability to enter animal body through skin penetration, peroral administration and pulmonary absorption beside initiative injection. The potential mechanism of fullerene toxicity is to interrupt the activation process of protein, cause mitochondrial dysfunction and generate ROS to damage organism. Nevertheless, the experiments result shows the negative effects of fullerenes only occur in a high dose and the assessments of fullerenes toxicity show a low result with the IC50 value and LD50 value to be 383.4 lg/mL to HEK293 and 721mg/kg to mice. It is suggested that fullerenes can rarely affect human health as long as people are exposed to the appropriate dose of fullerenes and the toxicity of fullerenes can further reduce via synthesis of fullerene derivates.

The cisplatin encapsulation into carbon nanohorns (CNH) is a promising nanoformulation to circumvent the drug dissipation and to specifically accumulate it in tumor sites. Herein, biased molecular dynamics simulations were used to analyze the transmembrane transport of the CNH loaded with cisplatin through a breast cancer cell membrane prototype. The simulations revealed a four-stage mechanism: approach, insertion, permeation, and internalization. Despite the lowest structural disturbance of the membrane provided by the nanocarrier, the average free energy barrier for the translocation was 55.2 kcal mol −1 , suggesting that the passive process is kinetically unfavorable. In contrast, the free energy profiles revealed potential wells of −6.8 kcal mol −1 along the insertion stage in the polar heads region of the membrane, which might enhance the retention of the drug in tumor sites; therefore, the most likely cisplatin delivery mechanism should involve the adsorption and retention of CNH on the surface of cancer cells, allowing the loaded cisplatin be slowly released and passively transported through the cell membrane.
