Study of HLA class I, class II and complement genes (C2, C4A, C4B and BF) in Japanese psoriatics and analysis of a newly-found high-risk haplotype by pulsed field gel electrophoresis

Abstract: Genetic polymorphisms of HLA antigens and HLA-linked serum complement components (C2, C4A, C4B and BF) were investigated in 79 Japanese patients suffering from psoriasis. HLA typing revealed increased frequencies of HLA-A1, A2, B39, Bw46, Cw6, Cw7 and Cw11. Among complement components, positive associations were obtained with C4A4 and C4B2 and a negative association with BFF. The major histocompatibility complex haplotype (supratype), HLA-A2-Cw11-Bw46-C2C-BFS-C4A4-C4B2-DRw8 is purported to be a new high-risk h… Show more

“…In these patients, apparent GVHD was not detected by clinical and histopathological findings. In contrast, the donor and recipient in this report had no psoriatic disposition or specific HLA subtypes believed to increase the risk for psoriasis vulgaris (HLA Cw6, Cw7, DR7, A1, A2, B13, B17, B39, Bw46 and Bw57) [8,9,10]. Interestingly, histological characteristics of both psoriasis and acute GVHD coexisted in the same specimen.…”

“…The class I HLA antigens A1, B13, B17, B37, B39, Cw6, Cw11 and class II HLA antigen DR7 occur with an increased frequency in patients with psoriasis [3,10,14,15,17,19,27,29] but the findings vary between patients of different racial and ethnic backgrounds [6,14,17,18,27]. The strong association of certain HLA genes, particularly Cw6, with psoriasis in most ethnic groups suggests that the genetic factors affecting the initiation or regulation of the immune response may be integrally linked with the pathogenesis of psoriasis.…”

“…In Japanese psoriatics, we have previously demon strated significant increases in the frequencies of HLA-A 1, A2, B39, Bw46, Cw6, Cw7 and C w ll [5].…”

“…The relationship of HLA-A, -B, -C and -DR antigens with psoriasis has been extensively studied. HLA-B13, -B17, -B37, -Cw6 and -DR7 were reported to be increased in frequency in Caucasian psoriatics [2][3][4], We have previously demonstrated that HLA-A 1, -A2, -B39, -Bw46, -Cw6, -Cw7 and -Cwll showed signif icantly increased frequencies in Japanese patients with psoriasis [5], Among these antigens, Cw6 is known to be increased in frequency in psoriatic patients regardless of their ethnic and racial backgrounds.…”
