Increased demand for livestock products is leading to research on more productive pastures adapted to different environments. Urochloa humidicola is a species adapted to low-fertility conditions and to environments occasionally subjected to temporary flooding and with highly dormant seeds at harvest. Mechanical and sulphuric acid (H2SO4) scarification are used to release these seeds from dormancy; although, there are several side effects. Hydrogen peroxide (H2O2) is linked to dormancy release in many crop species. The objective of this research was to characterize the dormancy release and H2O2 accumulation during one year of storage in dormant spikelets of U. humidicola cv. Llanero equilibrated to 4.5% and 50% relative humidity (RH) and stored at 20 °C, during which time the seeds had moisture contents (MCs) of 0.03 and 0.1 g H2O g-1 dry weight (DW), respectively. The seeds were evaluated initially and at 3, 6 and 12 months; the seeds from spikelets or just caryopses were evaluated. Germination and dormancy percentage, H2O2 content, superoxide dismutase (SOD) and peroxidase (PRX) activity were determined. The germination and H2O2 content increased mainly in the caryopses, and SOD activity increased. Moreover, dormancy and PRX activity decreased during storage. For the first time, it was shown, in U. humidicola cv. Llanero that dormancy release during storage is caused mainly by the accumulation of reactive oxygen species (ROS) in the spikelets even under ultra-dry storage conditions (4.5% RH), and this process could be accelerated by seed storage at a MC of 0.1 g H2O g-1 DW at 20 °C.

Current colorimetric methods for quantitative determination of seed viability (SV) with 2,3,5-triphenyl tetrazolium chloride (TTC) have been plagued by issues of being cumbersome and time-consuming during the experimental process, slow in extraction and staining, and exhibiting inconsistent results. In this work, we introduced a new approach that combines TTC-staining with high-temperature extraction using dimethyl sulfoxide (DMSO). The optimization of the germination stage, TTC-staining method, and 1,3,5-triphenylformazan (TTF) extraction method were meticulously carried out as follows: When the majority of wheat seeds had grown the radicle, and the length of radicles was approximately equal to the seed length (24 h-germination), 2 g germinating seeds were placed into a beaker (20 mL) containing 5 mL 10 g·L−1 TTC solution. The seeds were stained with TTC in the dark at 25 °C for 1 h. Following the staining, 1 mL 1 mol·L−1 H2SO4 was added to stop the reaction for 5 min. The H2SO4 solution was then removed, and the seeds were gently rinsed with deionized water. Subsequently, the TTF produced in the seeds was extracted directly with 5 mL DMSO solution at 55 °C for 1 h. The absorbance of the extract was measured at 483 nm, and the index of SV was calculated according to a predetermined TTC calibration curve and expressed by mg TTC·g−1 (seed)·h−1. The new method has been demonstrated to be rapid, stable, and highly sensitive, as evidenced by the accurate measurement of seed viability with different aging degrees.

The seed coat takes an important function in the life cycle of plants, especially seed growth and development. It promotes the accumulation of nutrients inside the seed and protects the seed embryo from mechanical damage. Seed coat permeability is an important characteristic of seeds, which not only affects seed germination, but also hinders the detection of seed vigor by electrical conductivity (EC) method. This research aimed to elucidate the mechanism of seed coat permeability formation through metabolome and transcriptome analysis of Elymus nutans. We collected the samples at 8, 18, and 28 days post-anthesis (dpa), and conducted a seed inclusion exosmosis experiment and observed the seed coat permeability. Moreover, we analyzed the changes in the metabolome and transcriptome during different development stages. Here, taking 8 dpa as control, 252 upregulated and 157 downregulated differentially expressed metabolites (DEMs) were observed and 886 upregulated unigenes and 1170 downregulated unigenes were identified at 18 dpa, while 4907 upregulated unigenes and 8561 downregulated unigenes were identified at 28 dpa. Meanwhile, we observed the components of ABC transporters, the biosynthesis of unsaturated fatty acids, and phenylalanine metabolism pathways. The key metabolites and genes affecting seed coat permeability were thiamine and salicylic acid. Furthermore, there were 13 and 14 genes with correlation coefficients greater than 0.8 with two key metabolites, respectively, and the —log2Fold Change— of these genes were greater than 1 at different development stages. Meanwhile, pathogenesis-related protein 1 and phenylalanine ammonia-lyase play an important role in regulating the formation of compounds. Our results outline a framework for understanding the development changes during seed growth of E. nutans and provide insights into the traits of seed coat permeability and supply a great significance value to seed production and quality evaluation.
