Succinamide Derivatives Ameliorate Neuroinflammation and Oxidative Stress in Scopolamine-Induced Neurodegeneration

Abstract: Oxidative stress-mediated neuroinflammatory events are the hallmark of neurodegenerative diseases. The current study aimed to synthesize a series of novel succinamide derivatives and to further investigate the neuroprotective potential of these compounds against scopolamine-induced neuronal injury by in silico, morphological, and biochemical approaches. The characterization of all the succinamide derivatives was carried out spectroscopically via proton NMR (1H-NMR), FTIR and elemental analysis. Further in vivo… Show more

“…Next, after the addition of 75 μL of thiobarbituric acid (TBA) to the mixture, the color changed, and the absorbance of the resultant mixture was immediately measured using a microplate reader (at 532 nm). The levels were expressed as nmol Tbras/min/mg protein ( Iqbal et al, 2020 ).…”

“…Lipid peroxidation assay was carried out by measuring thiobarbituric acid reactive substances (TBARS) using colorimetry, as described previously with slight modifications [ 33 ]. The assay mixture consists of 580 µL of 0.1 M phosphate buffer (pH 7.4), 200 µL supernatant, 20 µL of ferric chloride, 200 µL of 100 mM ascorbic acid and incubated on a water bath at 37 °C for 60 min.…”

“…Immunohistochemical staining was performed as previously described with minor modifications (Shah et al, 2019c;Iqbal et al, 2020). The slides were processed for the antigen retrieval step (enzymatic method), then washed with PBS.…”
