SUMOylation of the m6A-RNA methyltransferase METTL3 modulates its function

Abstract: The methyltransferase like 3 (METTL3) is a key component of the large N6-adenosine-methyltransferase complex in mammalian responsible for N6-methyladenosine (m6A) modification in diverse RNAs including mRNA, tRNA, rRNA, small nuclear RNA, microRNA precursor and long non-coding RNA. However, the characteristics of METTL3 in activation and post-translational modification (PTM) is seldom understood. Here we find that METTL3 is modified by SUMO1 mainly at lysine residues K177, K211, K212 and K215, which can be red… Show more

“…N6-methyladenosine (m6A) of RNA has been proposed as a new layer of epigenetic regulation. This biochemical process is proved to play important roles in regulation of cell growth, differentiation, and self-renewal through controlling RNA splicing, translation and stability [23,26,27]. Previous studies identified m6A as a predominant modification of mRNAs [7,23].…”

“…One line of evidence shows that posttranslational modification of FTO by ubiquitination regulates its proteasomal degradation, nuclear translocation, and demethylase activity (Tai et al 2017;Zhu et al 2018). More recently, SUMOylation of METTL3 has also been shown to regulate its methyltransferase activity (Du et al 2018).…”

“…It's interesting that the size of METTL3 is about 100 kD in all of the samples and is larger than predicted. This result indicated that METTL3 in silkworm may be modified by SUMOylation (Du et al, ).…”
