Sumoylation of transcription factor MYB30 by the small ubiquitin-like modifier E3 ligase SIZ1 mediates abscisic acid response in Arabidopsis thaliana

Abstract: The phytohormone abscisic acid (ABA) plays an essential role in plant development and during the response of the plant to abiotic stress. In this study, we report that the R2R3-type transcription factor MYB30 is involved in the regulation of ABA signaling. Arabidopsis mutants lacking MYB30 are hypersensitive to ABA during germination and seedling growth. A K283R substitution in MYB30 blocks its SUMO E3 ligase SIZ1-mediated sumoylation in Arabidopsis protoplasts, indicating that MYB30 is sumoylated by SIZ1 and … Show more

“…This constitutively expressing 35S:AEQUORIN line was used as the wild type in this study, and a T-DNA insertional mutant pool was constructed based on this wild type (Zhao et al, 2001(Zhao et al, , 2011Zheng et al, 2012). We screened for mutants with altered [Ca 2+ ] cyt when plants were treated with NaCl.…”

“…ABA is known to induce ROS cellular level in guard cells and the mitochondria at the root tip [12,15]. In addition, MYB30 is reported to be involved in seed germination under ABA signaling [17]. Moreover, MYB30 protein levels are stabilized by a SUMO E3 ligase, SIZ1, and this stabilization is important in regulating the expression of certain ABA response genes [18].…”

“…Col-0 was used as the wild type. The som-2 (salk_090314; Kim et al, 2008), som-3 (salk_008075; Kim et al, 2008), aba2-1 (Oh et al, 2009), ga1 (salk_109115;Oh et al, 2006), abi3-sk11 (salk_023411;Park et al, 2011), abi3-sk22 (salk_138922, also known as S138922; Kotak et al, 2007), and abi5-8 (salk_013163;Zheng et al, 2012) mutants were obtained from the Arabidopsis Stock Centers (Alonso et al, 2003), and all of them are in the Col-0 background. The aba2 ga1 double mutant was generated by crossing the aba2-1 and ga1 mutants.…”
