Suppression of 6A8 α‐mannosidase gene expression reduced the potentiality of growth and metastasis of human nasopharyngeal carcinoma

Abstract: Suppression of ␣-mannosidases by chemicals has been shown to reduce the potentiality of growth and metastasis of various tumors. In our study, the effect of 6A8 ␣-mannosidase (MAN 6A8), recently discovered in our laboratory, on malignant behaviors of tumor cells was examined. Since the suppressive effect of chemicals on ␣-mannosidase is not specific, antisense technique was used to specifically inhibit expression of the MAN 6A8 in human nasopharyngeal carcinoma cells, CNE-2L2. Key words: ␣-mannosidase; tumor;… Show more

“…This anti-apoptotic function of Man2C1 was shown to be independent of ␣-mannosidase activity; together, these findings provide convincing evidence that Man2C1 has a role in addition to that as a catabolic enzyme. Man2C1 up-regulation is reportedly involved in tumorigenesis of various cancer types; therefore, this protein has potential as an attractive target for anti-cancer therapy (16,19). Further clarification of the mechanism by which Man2C1 down-regulation induces apoptosis will provide clues for designing Man2C1-based anti-cancer reagents in the future.…”

“…These findings led us to speculate that this protein is a potential target for anti-cancer therapy. Suppression of the Man2C1 gene was shown to induce growth delay or arrest in nasopharyngeal carcinoma cells (19) and in Hek293 cells (12). Down-regulation of Man2C1 reportedly also results in mitotic arrest and apoptosis in esophageal carcinoma cells (21).…”

“…Moreover, up-regulation of MAN2C1 was observed in human prostate cancer cells, where MAN2C1 seemed to attenuate PTEN functions by binding to PTEN (19). Conversely, down-regulation of MAN2C1 induced growth delay or arrest in nasopharyngeal carcinoma cells (41) and led to mitotic arrest and apoptosis in esophageal cancer cells (20). Recently, the antiapoptotic function of MAN2C1 was investigated.…”
