Suppression of microRNA activity amplifies IFN-γ-induced macrophage activation and promotes anti-tumour immunity

Abstract: Tumour-associated macrophages (TAMs) largely express an alternatively activated (or M2) phenotype, which entails immunosuppressive and tumour-promoting capabilities. Reprogramming TAMs towards a classically activated (M1) phenotype may thwart tumour-associated immunosuppression and unleash anti-tumour immunity. Here we show that conditional deletion of the microRNA (miRNA)-processing enzyme DICER in macrophages prompts M1-like TAM programming, characterized by hyperactive IFN-γ/STAT1 signalling. This rewiring … Show more

“…Macrophage‐specific Dicer1 deletion significantly delayed tumor progression. They demonstrate that let‐7‐5p family members were upregulated in Dicer‐knockout and wild‐type TAMs (Baer et al ., 2016). These findings suggest that Dicer1 deletion can regulate the antitumor phenotype of M2 TAMs to M1‐like TAMs in different types of tumor models (Baer et al ., 2016).…”

“…Conversely, macrophages' iron controls the production of cytokines by these cells . Furthermore, it has been shown that IFNγ stimulates M1 activation resulting in decreased immune activity of M2 cells which are tumor associated macrophages (Baer et al, 2016). Jason et al (2001) have reported that iron deficiency in children is associated with higher percentage of lymphocytes producing IL-6, IFNγ and IL-4.…”

“…Global ablation of miRNAs by deletion of Dicer1 in mice resulted in polarisation of M2 towards M1 population and boosted antitumour reactivity. (15) This process was rescued by forced expression of let-7 miRNA, (15) suggesting that the tumourtolerant M2 phenotype is chiefly maintained by let-7 miRNA.…”
