Suppressive Effects of Mesenchymal Stem Cells in Adipose Tissue on Allergic Contact Dermatitis

Abstract: BackgroundAllergic contact dermatitis (ACD), which is accelerated by interferon (IFN)-γ and suppressed by interleukin (IL)-10 as regulators, is generally self-limited after removal of the contact allergen. Adipose tissue-derived multipotent mesenchymal stem cells (ASCs) potentially exert immunomodulatory effects. Considering that subcutaneous adipose tissue is located close to the site of ACD and includes mesenchymal stem cells (MSCs), the MSCs in adipose tissue could contribute to the self-limiting course of … Show more

“…Transforming growth factor (TGF-β1) production from hUC-MSCs in response to interleukin-4 (IL-4) contributes to the attenuation of MC degranulation by downregulating Fc𝜀RI expression in MCs ( Kim et al, 2015 ). Adipose tissue-derived mesenchymal stem cells (AdMSCs) contribute to the self-limiting course of ACD by decreasing the expression of interferon-γ (IFN-γ) ( Kikuchi et al, 2017 ). They can reduce gross and histological signatures of AD induced by Dermatophagoides farinae , decrease serum IgE level, and suppress MC degranulation ( Shin et al, 2017 ).…”

“…These models involve an initial application of chemical sensitiser, followed by a challenge with the same chemical several days later to elicit a robust T cellmediated inflammatory response featuring immune cell recruitment and swelling (Deane & Hickey 2009, Deane et al 2012. MSC from bone marrow (Chen et al 2018), adipose tissue (Kikuchi et al 2017) and gingiva (Su et al 2011, Li et al 2016 have the ability to inhibit ear swelling in contact sensitivity when delivered via and intravenous route. In contrast, eMSCs did not inhibit ear swelling in an oxazolone-induced model of contact sensitivity.…”

“…105 Human ADSC-derived exosomes demonstrated strong immunomodulatory effects in preclinical AD murine model. 106,107 Th2 cytokines (TNFα, IL-4, IL-13, IL-23 and IL-31) in skin lesions resulted reduced after ADSC-derived exosomes intravenous injection or subcutaneous administration. 103 Notably, at higher doses, the observed effect was fully comparable with that of oral prednisolone.…”
