Surface‐activated chemical ionization time‐of‐flight mass spectrometry and labeling‐free approach: two powerful tools for the analysis of complex plant functional proteome profiles

Abstract: Surface-activated chemical ionization (SACI) has been widely used in recent years to analyze a range of different compounds (e.g., peptides, street drugs, amino acids). The main benefits of this technology are its high sensitivity and its effectiveness under different chromatographic conditions. Here, we used SACI in conjunction with a highly selective quadrupole time-of-flight mass analyzer to characterize a complex proteome pattern after separation by sodium dodecyl sulfate polyacrylamide gel electrophoresis… Show more

“…Immunoprecipitates were eluted, protein complexes were subjected to proteolytic digestion, and peptide mixtures were analyzed by mass spectrometry at Ion Source & Biotechnologies S.r.l. (Milan) as previously described ( 46 ). Mass spectra were obtained using a q-TOF mass analyzer (Bruker Daltonics, Billerica, MA, USA).…”

“…Orbitrap, QTOF etc). Very recently SACI technology has been applied in seagrass proteomics (Finiguerra et al, 2010). In fact, the increasing sensitivity of this ionization device improves peptides detection thus recovering the limited sea grass genome resources.…”

“…The SACI technique strongly increased the number of detectable proteins and of assigned peptides for each protein. For example, with SACI technology application, it was possible to identify a previously identified protein (a heat shock cognate protein), 1000 fold over expressed in deeper plants (-27 m) in comparison with the more shallow plants (-5m), detecting four peptides respect to only two detected by micro-ESI (Finiguerra et al, 2010). Fig.…”

“…This large amount of information should be added to relevant seagrass databases in order to facilitate the organization of data to generate testable hypothesis. The application of new technique(s) combining two- or one-dimensional (1D) SDS-PAGE with a high-mass-accuracy LC-ESI-MS and LC-SACI-MS and MS/MS to sequence identification approaches has demonstrated an increase in the confidence of results (Finiguerra et al, 2010). This can provide a high-throughput system to achieve the goal of sequencing complete proteomes from seagrass organs and tissues.…”
