Surface antigens of rat liver epithelial cells grown in medium containing foetal bovine serum

Abstract: Summary.-Cultured rat liver cells induced a strong antibody response in syngeneic rats, directed against foetal calf serum components which were incorporated into the liver cell surface from the cell-culture medium. This antibody could be removed by absorption with liver cells or glutaraldehyde-fixed foetal calf serum. It is possible that the antigenic cross-reactivity observed in earlier studies with cultured cells treated with carcinogens could be due to this foetal calf serum component.

“…Additionally, antigenic determinants were expressed on transplanted respiratory-tract carcinoma induced in vivo with DMBA and not adapted to cell culture. Thus, growth in tissue culture is not required for antigenic expression as has been demonstrated for other cell culture systems (Thorpe et al, 1977;Peterson et al, 1981) or due to media components non-specifically absorbed to the cell surface (Embleton and Iype, 1978).…”

“…F-344 respiratory tract cell lines do not, however, exhibit significant reverse transcriptase activity, nor is there antibody in immune sera for p30 or gp70 molecules of murine leukemia virus (Braslawsky et al, 1981;Jamasbi and Nettesheim, 1977). Immunization with cells propagated in v i m does pose the possibility of antibody responses against tissue culture components (Embleton and Iype, 1978). For this reason, all immunizations were done using cultures propagated in syngeneic rat serum.…”

“…Tumorigenicity of the MNNG-induced tracheal cell lines was determined by inoculation at various passages of 106 cells into immunosuppressed F-334 rats or BALB/c (nu/nu) mice (Table I) culture can adsorb antigenic determinants from the serum in culture medium (Embleton & Iype, 1978) In order to test more fully for the presence of antigen-bearing cells among normal or transformed epithelial cells (Engers & MacDonald, 1976) immune spleen cells were co-cultivated for 6 days in vitro with primary tracheal-outgrowth cultures or neoplastic epithelial cell lines before 20h microcytotoxicity testing. As shown in Table IV, significant reactivity for the immunizing target cell was obtained when immune spleen cells were co-cultivated with either of the neoplastic cell lines.…”
