Survey of Deoxynivalenol Contamination in Agricultural Products in the Chinese Market Using An ELISA Kit

Abstract: A total of 328 agricultural product samples highly suspected to be contaminated, from flour companies, feed companies, and livestock farms throughout China, were surveyed for deoxynivalenol (DON) contamination using a self-assembly enzyme-linked immunosorbent assay (ELISA) kit. An ELISA kit for DON was developed with a 4.9 ng mL−1 limit of detection (LOD) in working buffer and a 200 ng g−1 LOD in authentic samples. The DON contamination detection rate was 88.7%, concentrations ranged from 200.9 to 6480.6 ng g−… Show more

“…Compared with the commercial kits, the sensitivity and specificity is higher (3 ng/mL in the working buffer), the DON levels of samples were equivalent to a 100-fold dilution with an LOD of 300 ng/g in authentic agricultural samples, and the cross-reaction rate with 3-Ac-DON and 15-Ac-DON was less than 70%, less than 1%, respectively. Compared with the kit that was developed by Li et al [12], it has higher sensitivity and specificity (4.9 ng/mL in the working buffer), the DON levels of samples were equivalent to a 40-fold dilution with an LOD of 200 ng/g in authentic agricultural samples, and the cross-reaction rate with 3-Ac-DON and 15-Ac-DON was 5.7%, less than 0.5%, respectively. Accuracy and precision are measured by spiked sample recovery (%) and relative standard deviation (RSD%).…”

“…In view of its serious toxic effects, in the preliminary draft of the DON maximum levels (MLs), the Codex Alimentarius Food (FAO) Committee recommended the following limits: 2 mg/kg in unprocessed cereals, 1 mg/kg in semi-processed products using wheat, corn and barley as raw materials, and 0.5 mg/kg in cereals for infants and young children [10,11]. In China, the ML of DON in maize, wheat, and their products is regulated at 1 mg/kg [12,13]. The molecular formula of DON is C15H20O6, and its molecular weight is 296.32 [14].…”

“…Studies to determine the mycotoxin content of food products produced in different countries and sold in markets have accelerated all over the world (Reddy et al, 2009;Houissa et al, 2019;Potortì et al, 2020;Costa et al, 2019). Li, Sun, Hong, Duan, and Du (2019) detected 88.7% contamination in their DON study on 328 agricultural products with ELISA, ranging from 0.2009 to 6.4806 μg kg −1 . In another study, El-Shanshoury, El-Sabbagh, Emara, and Saba (2014) investigated the AFT and AFB 1 values of cereal grains and peanut sold in central delta provinces markets, Egypt by using thin layer chromatography method.…”

“…and mainly contaminates wheat, maize and other crops in the field and during storage ( Mishra et al, 2019 ). DON seriously threatens the health of humans and animals, as it shows enterotoxicity, immunotoxicity and teratogenicity ( Li et al, 2018a ; Liao et al, 2018 ). Pigs are the most sensitive species to DON and intestine is the most important target organ of DON which could induce intestinal inflammation and disrupt the intestinal barrier function ( Alassane-Kpembi et al, 2017 ; Ananthakrishnan et al, 2018 ).…”
