Susceptibilities of Haemophilus influenzae and Moraxella catarrhalis to ABT-773 Compared to Their Susceptibilities to 11 Other Agents

Abstract: The activity of the ketolide ABT-773 against Haemophilus and Moraxella was compared to those of 11 other agents. Against 210 Haemophilus influenzae strains (39.0% ␤-lactamase positive), microbroth dilution tests showed that azithromycin and ABT-773 had the lowest MICs (0.5 to 4.0 and 1.0 to 8.0 g/ml, respectively), followed by clarithromycin and roxithromycin (4.0 to >32.0 g/ml). Of the ␤-lactams, ceftriaxone had the lowest MICs (<0.004 to 0.016 g/ml), followed by cefixime and cefpodoxime (0.008 to 0.125 and <… Show more

“…The PAEs produced by the novel macrolides, telithromycin, and azithromycin were longer than those produced by erythromycin. These results are consistent with those from previous studies (3,15). The PAE would be important only for organisms for which the MICs are high and in cases in which the levels in serum would fall below the MIC.…”

“…Credito et al determined the PAEs of ABT-773 and 11 other antibiotics against five strains of H. influenzae (5). Unlike the previously mentioned study, the antibiotic concentrations varied based upon achievable pharmacokinetic parameters.…”

“…The incidence of ␤-lactamase-negative ampicillin-resistant (BLNAR) strains is currently Ͻ1% in the United States (11) but is significantly higher in Japan (23). Among members of the macrolide and azalide groups, azithromycin has the lowest MICs against these organisms, followed by ketolides such as telithromycin, erythromycin A, and clarithromycin (1,5,6,10,14,17). Of available streptogramins, oral pristinamycin and parenteral quinupristin/ dalfopristin have MICs for H. influenzae and H. parainfluenzae which are several dilutions higher than those for pneumococci (8,12,15).…”

“…The major bacterial pathogens responsible for communityacquired respiratory tract infections comprise Streptococcus pneumoniae, Haemophilus influenzae, and Moraxella catarrhalis (6,7,(9)(10)(11)24). Pneumococcal resistance to penicillin G and other ␤-lactam and non-␤-lactam compounds has increased worldwide at an alarming rate, including in the United States.…”
