Susceptibility of HepG2 Cells to Silver Nanoparticles in Combination with other Metal/Metal Oxide Nanoparticles

Abstract: The fast-growing use of nanomaterials in everyday life raises the question about the safety of their use. Unfortunately, the risks associated with the use of nanoparticles (NPs) have not yet been fully assessed. The majority of studies conducted so far at the molecular and cellular level have focused on a single-type exposure, assuming that NPs act as the only factor. In the natural environment, however, we are likely exposed to a mixture of nanoparticles, whose interactions may modulate their impact on living… Show more

“…In the in vivo experiment, the studied animals were exposed to AgNPs with three different types of surface coating. AgNPs with bovine serum albumin (BSA; PlasmaChem, Berlin, Germany) coating and a nominal diameter of 20 ± 5 nm were prepared as described previously [65,66]. Briefly, 2 mg of AgNPs was dispersed in 800 µL of purified distilled water to form the nanoparticle stock solution.…”

“…The well-differentiated hepatoma cell line HepG2, which has a wide set of properties inherent to human hepatocytes in vivo, has been actively used for about 40 years in various studies exploiting organ-specific features of the line [ 10 , 11 , 34 , 35 , 36 , 37 ]. Morphology of HepG2 cells was shown to keep main features of the hepatocytes: tight junctions separate apical and basolateral plasmalemma providing formation of bile capillaries and blood-biliary barrier; ultrastructural observations were supported by immunohistochemical and biochemical studies [ 38 , 39 ].…”

“…AgNPs with a of nominal diameter of 20 ± 5 nm were acquired from PlasmaChem (Berlin, Germany). Nanoparticles preparation was carried out as described previously [ 54 ]. Briefly, a stock solution of nanoparticles was prepared by dispersing 2 mg of AgNPs in 800 μL of distilled water.…”
